目的 评价百合乌药蒲公英汤(decotion of Baihe Wuyao Pugongying, DBWP)对肝纤维化(liver fibrosis, LF)的保护作用, 并揭示其相关机制。方法 通过四氯化碳诱导6~8周龄雄性昆明种小鼠LF模型并随机分为5组, 给予6周不同剂量DBWP进行体内实验验证。检测肝功能指标、肝脏病理形态和纤维化沉积; Western-blot检测胶原纤维因子及细胞外基质(extracellular matrixs, ECMs)降解酶表达, 反转录聚合酶链反应(reverse transcription-polymerase chain reaction, PT-PCR)检测炎症相关因子表达。结果 与模型组相比较, DBWP可明显改善肝功能, 降低小鼠血清谷丙转氨酶(alanine amino transferase, ALT)和谷草转氨酶(aspartate amino transferase, AST)水平。HE染色和Masson染色表明, DBWP可改善小鼠肝脏病理形态和纤维化沉积, 同时, 肝脏中Collagen 1和Collagen 3表达明显降低, 纤维化明显改善。基质金属蛋白酶(matrix metalloproteinases, MMPs)/金属蛋白酶抑制因子1 (metalloproteinase 1, TIMP1)比值明显升高, 肝脏对细胞外基质降解能力进一步加强。炎症相关因子白细胞介素-1β (interleukin-1β, IL-1β)和白细胞介素-6 (interleukin-6, IL-6)的表达明显降低。结论 DBWP通过抗炎、促进细胞外基质降解改善肝纤维化。 |

Objective To evaluate the protective effects of the decoction of Baihe Wuyao Pugongying (DBWP) on liver fibrosis (LF), and reveal the underlying mechanisms. Methods LF models of 6-8 week-old male Kunming mice were induced by carbon tetrachloride and randomly divided into 5 groups, which were given DBWP at different doses for 6 weeks for in vivo experimental validation. Liver function index, liver pathological morphology and fibrosis deposition were detected. The expression of collagen and extracellular matrixs (ECMs) degrading enzymes was detected by Western-blot. The expression of inflammation-related factors was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results Compared with model group, DBWP significantly improved liver function and decreased serum aspartate amino transferase (ALT) and aspartate amino transferase (AST) levels. HE staining and Masson staining showed that DBWP could improve the pathological morphology and fibrosis deposition of mice liver. Meanwhile, the expressions of Collagen 1 and Collagen 3 in the liver were decreased, and the fibrosis was significantly improved. The ratio of matrix metalloproteinases (MMPs)/metalloproteinase 1 (TIMP1) increased, and the ability of liver to degrade extracellular matrix was further enhanced. The expression of inflammation-related factors interleukin-1β (IL-1β) and interleukin-6 (IL-6) was significantly decreased. Conclusion DBWP can alleviate LF mainly through anti-inflammatory and promoting the degradation of extracellular matrix. |
