      目的 建立高效液相色谱法快速检测婴幼儿配方食品中维生素B12的分析方法。方法 选择C18柱(250 mm×4.6 mm, 5.0 μm)作为色谱柱, 流动相为水+甲醇, 流速0.5 mL/min等度洗脱, 进样量35 μL, 柱温40 ℃, 波长361 nm, 样品经超声提取15 min, 后在8000 r/min下冷冻离心10 min, 离心结束后取上清液过 0.22 μm滤膜, 上机测试。结果 维生素B12标准品在浓度0.05~2.0 μg/mL范围内线性良好; 相关系数r为0.9998; 平均回收率为91.3%; 检出限为2 ng/mL。结论 该方法重复性好、简单、快速、结果准确, 可应用于婴幼儿配方食品中维生素B12含量的检测。
英文摘要:
      Objective To establish a method for rapid detection of vitamin B12 in infant formula foods by high performance liquid chromatography. Methods C18 column (250 mm×4.6 mm, 5.0 μm) was selected as the chromatographic column, the mobile phase was water+methanol (25%), the flow rate was 0.5 mL/min using isocratic elution, the injection volume was 35 μL, the column temperature was 40 ℃, and the wavelength was 361 nm. The sample was extracted by ultrasonic for 15 min, and then centrifuged at 8000 r/min for 10 min. After centrifugation, the supernatant was taken through a 0.22 μm filter membrane and tested on the machine. Results The standard vitamin B12 had good linearity within the concentration range of 0.05?2.0 μg/mL. The correlation coefficient r was 0.9998. The average recoveries was 91.3%, and the limit of detection was 2 ng/mL. Conclusion This method is repeatable, simple, rapid and accurate, whice can be applied to the determination of vitamin B12 in infant formula foods.
