目的 建立超高效液相色谱法(ultra performance liquid chromatography, UPLC)测定清咽含片中罗汉果皂苷V含量的方法。方法 色谱条件为色谱柱: XSELECT HSS T3 (100 mm×2.1 mm, 2.5 μm); 流动相: 乙腈: 0.1%磷酸水溶液, 梯度洗脱, 流速: 0.4 mL/min; 柱温: 40 ℃; 进样量: 5 μL; 检测波长: 203 nm。结果 结果表明, 罗汉果皂苷V线性范围为0.033~0.333 mg/mL(r=1.0000), 检出限66.4 μg/g, 定量限221.6 μg/g, 精密度(relative standard deviation, RSD)为1.5%, 平均回收率97.6%(n=9)。结论 此法操作简单、准确、快速, 可作为清咽含片中罗汉果皂苷V含量的检测方法。 |

Objective To establish a method for determination of the content of saponin V from Siraitia grosvenorii in Chinese pharynx tablets by ultra performance liquid chromatography (UPLC). Methods The chromatographic conditions were as follows: chromatographic column was XSELECT HSS T3 (100 mm×2.1 mm, 2.5 μm), flow phase was acetonitrile: 0.1% phosphoric acid water with gradient elution, flow rate was 0.4 mL/min, column temperature was 40 ℃, sample amount was 5 L, and detection wavelength was 203 nm. Results The results showed that the linear range of saponin V was 0.033~0.333 mg/mL (r=1.0000). The limit of detection was 66.4 g/g, and the limit of quantitative was 221.6 g/g, the relative standard deviation (RSD) was 1.5%, and the average recovery rate was 97.6% (n=9). Conclusion This method is simple, accurate and rapid, which can be used for determination of the content of saponin from Siraitia grosvenorii in Chinese pharynx tablets. |
