目的 建立高效液相色谱法(high performance chromatography, HPLC)测定营养强化剂中氯化高铁血红素的含量。方法 采用0.1 mol/L氢氧化钠溶液提取样品, 以Agilent TC-C18为色谱分析柱, 流动相为0.6%乙酸:甲醇=30:70(V:V), 流速1.0 mL/min, 柱温35 ℃, 检测波长399 nm, 采用高效液相色谱-二极管阵列检测器进行检测, 以峰面积外标法定量。结果 该方法在40~140 mg/L范围内线性关系良好, 相关系数r为0.9993, 相对标准偏差(relative standard deviation, RSD)小于1.38%。结论 该方法操作简单、快速、准确、灵敏度高, 适合测定营养强化剂中氯化高铁血红素的含量。 |

Objective To establish a method for determination of hemin in nutrient supplement by high performance chromatography (HPLC). Method The sample was extracted with 0.1 mol/L sodium hydroxide solution, and was analyzed with Agilent TC-C18. The mobile phase was 0.6% acetic acid: methanol=30:70 (V:V), the flow rate was 1.0 mL/min, and the column temperature was 35 ℃. The detection wavelength was 399 nm, and the detection was performed by a high performance liquid chromatography-diode array detector. And the samples were quantified by peak area external standard method. Result The method had a good linear relationship in the range of 40-140 mg/L, the correlation coefficient r was 0.9993, and the relative standard deviations (RSDs) were less than 1.38%. Conclusion The method is simple, rapid, accurate and sensitive, which is suitable for the determination of hemin in nutrient supplement. |
