      目的 制备新型立体结构色谱固定相并开发新的高效液相色谱法(high performance liquid chromatography, HPLC)分离7种有机酸的方法。方法 采用新型立体结构固定相对7种有机酸进行分离, 以5%甲醇-95% 10 mmol/L甲酸铵(pH=3.0)水溶液作为流动相进行等度洗脱; 流速为1.0 mL/min; 柱温: 40 ℃; 紫外检测波长: 220 nm; 进样体积: 10 μL, 采用HPLC进行检测。结果 制备了新型立体结构固定相, 具有与常规C18固定相相似的疏水作用, 8 min内完全分离7种有机酸, 且分离效果较好。结论 本方法具有保留增强、快速及分离效果好等优势, 可用于多种有机酸的分离测定。
英文摘要:
      Objective To prepare a new solid structure stationary phase and establish a new method for separation of 7 organic acids by high performance liquid chromatography (HPLC). Methods Totally 7 organic acids were separated by new solid structure stationary phase with the 5% methanol-95% 10 mmol/L ammonium formate (pH=3.0) solution as a mobile phase for isocratic elution. Flow rate was 1.0 mL/min, column temperature was 40 ℃, detection wavelength was 220 nm and injection volume was 10 μL. Then the samples were detected by HPLC. Results A new solid structure stationary phase was prepared, which had hydrophobic effect similar to that of C18 stationary phase by new solid structure stationary phase. Totally 7 organic acids were completely separated within 8 min, and the separation effect was better. Conclusions The established method has the advantages of strong retention, rapid determination and good separation efficiency, which is suitable for detection of many organic acids.
