      目的 建立一种用于测定减肥类保健食品中番泻苷A、番泻苷B的高效液相色谱(high performance liquid chromatography, HPLC)检测方法。方法 采用ZORBAX SB-C18柱(4.6 mm×150 mm, 5 μm), 以乙腈-0.1%磷酸水溶液为流动相进行梯度洗脱, 流速为1.0 mL/min, 柱温为40 ℃, 二极管阵列检测器进行检测, 检测波长为254 nm。结果 番泻苷A、番泻苷B分别在2.050~20.50 μg/mL(r=0.9999)、1.803~18.03 μg/mL(r=0.9993)范围内呈良好线性关系, 定量限分别为30、27 μg/kg; 平均回收率分别为96.8%、95.1%, 相对标准偏差分别为1.3%、2.3%。结论 该方法简单、可行, 结果准确、可靠, 适用于减肥类保健食品中番泻苷A、番泻苷B含量的测定。

      Objective To establish a method for the determination of sennoside A and sennoside B in slimming health foods by high performance liquid chromatography (HPLC). Methods Samples were separated on a ZORBAX SB-C18 column (4.6 mm×150 mm, 5 μm). The mobile phase was composed of acetonitrile and 0.1% phosphoric acid with gradient elution. The flow rate was 1.0 mL/min and the column temperature was 40 ℃. The samples were detected with diode array detector, and the detection wavelength was 254 nm. Results Sennoside A and sennoside B had good linear relationships in the ranges of 2.050~20.50 μg/mL (r=0.9999) and 1.803~18.03 μg/mL (r=0.9993), respectively. The limits of quantification (LOQ) were 30 μg/kg and 27 μg/kg. The average recoveries were 96.8% and 95.1%, and the relative standard deviations (RSDs) were 1.3% and 2.3%. Conclusion The method is simple, specific and accurate, which is suitable for determination of sennoside A and sennoside B in slimming health foods.
