目的 在已制备出抗一类有机磷农药的组特异性单克隆抗体的基础上, 制备胶体金, 从而快速地对上述有机磷农药进行检测。方法 通过鞣酸-柠檬酸三钠还原法制备出直径16.08±0.64 nm、质地均一的胶体金, 并且以此标记单克隆抗体, 组装胶体金免疫层析检测板。利用紫外光谱、荧光光谱、透射电镜、动态光散射等方法对胶体金及金标抗体进行鉴定。结果 该检测板对标准毒死蜱的检测限为4 μg/mL, 检测时间为5 min。 结论 胶体金试纸条对毒死蜱具有较好的检测灵敏度, 并且基质的干扰效应不明显。 |

Objective To prepare the colloidal gold for the rapid detection of organicphosphorus pesticides on the basis of the specific monoclonal antibodies against a class of organicphosphorus pesticides. Methods Uniform texture colloidal gold with a diameter of 16.08+0.64 nm was prepared by tannin-sodium citrate reduction method, then labeled monoclonal antibody, and assembled colloidal gold immune chromato-graphy testing board. The colloidal gold and gold labeled antibodies were identified by ultraviolet spectrum, fluorescence spectroscopy, transmission electron microscopy, and dynamic light scattering. Results The testing board was validated to complete a detection assay within 5 min, and the LOD value of chlorpyriphos-ethyl was 4 μg/mL. Conclusion Colloidal gold test strip had a high sensitivity, and the matrix interference effect was not obvious. |
