ABSTRACT: Objective A method for simultaneously determining the content of linoleic acid and squalene in Hainan camellia oi was established by gas chromatography (GC). Methods In the test, 5mL 2mol/L KOH-methanol solution was added to the sample, and the vortex mixed 2min, 55°C ultrasonic (heating power 800W) water bath for 30 minutes to saponify the sample, and extracted with n-hexane. The DB-23 capillary column (60m × 0.250 mm × 0.25μm) was selected for chromatographic separation and analysis, and the hydrogen flame ionization detector was used for detection, and the external standard method was used for quantification.Results The results showed that the linear relationship between methyl linoleate in the range of 100~4000mg/L and squalene in the range of 10~400mg/L was good, and the correlation coefficient were greater than 0.999; the detection limit of linoleic acid is: 60mg/kg, squalene is 20mg/kg, the standard recovery rate range of the two compounds is: 90.9%-98.5%, and the relative standard deviation (RSD) was in the range of 0.5%-1.7%,The relative deviation of the average value of the sample is in the range of 3.2%-7.0% by comparison between different methods.Conclusion The method has high accuracy and good repeatability, and is suitable for the simultaneous determination of the content of linoleic acid and squalene in camellia oil. |
