目的 探究β-烟酰胺单核苷酸(NMN)对高脂血症模型大鼠的降脂和对肝脏的保护作用。方法 50只雄性SD大鼠,随机分为正常组、模型组和β-烟酰胺单核苷酸低、中、高剂量组(10mg/kg,50mg/kg,250mg/kg),每组10只,正常组给予维持饲料,其余各组给予高脂饲料建立高脂模型,造模成功后分别进行灌胃干预。干预5周后,检测血清生化指标。血脂,总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-c)及高密度脂蛋白(HDL-c)含量;血糖(GLU);肝肾功能;碱性磷酸酶(ALP)、丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、肌酐(CREA)水平,并结合病理观察肝脏组织学变化和进行NAS评分。 结果 与正常组相比,模型组大鼠体重增加明显,其血清中的TG、TC、LDL-c、AST、ALT和ALP含量极显著上升(P<0.05,P<0.01),HDL-c含量显著降低(P<0.05),GLU和CREA无显著变化。与模型组相比,低中高剂量组TG、TC、LDL-c、ALT显著降低(P<0.05),P<0.01);高剂量组AST含量显著降低(P<0.05);HDL-c、ALP、GLU和CREA无显著差异。与模型组相比,低中高剂量组对大鼠的肝细胞脂肪变性,炎症浸润均有改善,中高剂量组的NAS总分显著降低(P<0.05,P<0.01)。结论 β-烟酰胺单核苷酸能降低高脂血症SD大鼠的血脂水平,并对肝脏有一定的保护作用。 |

Objective To investigate the lipid-lowering and liver protective effects of β - nicotinamide mononucleotide on hyperlipidemic model rats. Methods Fifty male SD rats were randomly divided into a normal group, a model group, and low, medium, and high dose groups of β - nicotinamide mononucleotide (10mg/kg, 50mg/kg, 250mg/kg), with 10 rats in each group. The normal group was given maintenance feed, while the other groups were given high-fat feed to establish a high-fat model. After successful modeling, gastric lavage intervention was performed in each group. After 5 weeks of intervention, serum biochemical indicators and blood lipids were measured, including total cholesterol (TC), triglycerides (TG), low-density lipoprotein (LDL-c), and high-density lipoprotein (HDL-c) levels; Blood glucose (GLU); Liver and kidney function: alkaline phosphatase (ALP), alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine (CREA) levels, combined with pathological observation of liver histological changes and NAS of liver tissues was evaluated. Results Compared with the normal group, the body weight of the model group rats increased significantly, and the levels of TG, TC, LDL-c, AST, ALT, and ALP (p<0.05, p<0.01) in their serum increased significantly. The level of HDL-c (p<0.05) decreased significantly, while GLU and CREA showed no significant changes. The TG, TC, LDL-c, and ALT levels in the low, medium, and high dose groups were significantly reduced (p<0.05, p<0.01); The AST content in the high-dose group was significantly reduced (p<0.05); There was no significant difference in HDL-c, ALP, GLU, and CREA. Compared with the model group, the low, medium, and high dose groups improved hepatic steatosis and inflammatory infiltration in rats, and the total score of NAS in the medium and high dose groups was significantly reduced. Conclusion β-nicotinamide mononucleotide can reduce the blood lipid levels in hyperlipidemic SD rats and has a certain protective effect on the liver. |
