目的:建立豆类食品中大豆凝集素的酶联免疫检测方法。方法:样品经洗净后,加入磷酸盐缓冲溶液充分匀浆后,经离心提纯后,收集上清液经酶联免疫法测定,用酶标仪定量。结果:在0~400μg/mL的线性范围内,所得大豆凝集素的线性方程为y=0.002X+0.0579,r=0.9998,该方法的检出限为125mg/kg,回收率在 99.17%~101.2%之间,相对标准偏差在1.57~3.26%。结论 该方法操作简便,特异性好,抗原浓度与OD450值之间呈正比。适用于基层疾控中心对大豆凝集素中毒事件的检测。 |

Objective To establish an enzyme-linked immunosorbent assay (ELISA) method for detecting soy agglutinin in legume foods. Method: After washing the sample, add phosphate buffer solution and homogenize it thoroughly. After centrifugation and purification, collect the supernatant and determine it by enzyme-linked immunosorbent assay. Quantify it using an enzyme-linked immunosorbent assay (ELISA) reader. Result: In the linear range of 0-400 μ g/mL, the linear equation of soy agglutinin obtained is y=0.002X+0.0579, r=0.9998. The detection limit of this method is 125mg/kg, the recovery rate is between 99.17% and 101.2%, and the relative standard deviation is between 1.57-3.26%. Conclusion: This method is easy to operate, has good specificity, and the antigen concentration is directly proportional to the OD450 value. Applicable to the detection of soybean agglutinin poisoning incidents by grassroots disease control centers. |
