目的 研究超声和硫酸铜(CuSO4)对金黄色葡萄球菌的协同抗菌效应, 并深入探究二者的协同抗菌机制。方法 通过菌落计数法观察超声和CuSO4单独和协同处理前后金黄色葡萄球菌的生长情况, 运用扫描电子显微镜、β-半乳糖苷酶试验、荧光探针法等检测细胞形态、细胞膜通透性、膜电位的变化, 分析超声和CuSO4的协同抗菌机制。结果 超声联合CuSO4处理在体外和小番茄表面均能够显著抑制金黄色葡萄球菌的生长(P<0.05), 菌落数明显下降。经二者协同处理后, 细菌细胞膜出现明显收缩和破裂, 细胞膜渗透性增大, 膜完整性被破坏, 细胞内核酸蛋白质渗漏, 膜电位发生超极化。结论 超声与CuSO4具有显著的协同抗菌效果, 通过破坏金黄色葡萄球菌细胞膜的完整性和通透性, 造成细胞内物质外泄, 促进铜离子进入细胞发挥抑菌效应, 从而达到协同杀菌的目的。 |

Objective To investigate the synergistic antibacterial effects of ultrasound and copper sulfate (CuSO4) on Staphylococcus aureus and to explore their synergistic antibacterial mechanisms in depth. Methods The growth of Staphylococcus aureus was observed using colony counting methods before and after treatment with ultrasound and CuSO4, both individually and in combination. Scanning electron microscopy, β-galactosidase assays, and fluorescent probe methods were employed to detect changes in cell morphology, cell membrane permeability, and membrane potential. The synergistic antibacterial mechanisms of ultrasound and CuSO4 were analysed. Results The ultrasound combined with CuSO4 treatment significantly inhibited the growth of Staphylococcus aureus both in vitro and on the surface of cherry tomatoes, with a noticeable decrease in colony counts. After the combined treatment, the bacterial cell membranes showed significant shrinkage and rupture, permeability increased, membrane integrity was compromised, intracellular nucleic acids and proteins leaked out, and membrane potential hyperpolarization occurred. Conclusion There is a significant synergistic antibacterial effect between ultrasound and CuSO4. This effect is achieved by disrupting the integrity and permeability of the cell membrane of Staphylococcus aureus, leading to the leakage of intracellular substances and promoting the entry of copper ions into the cells to exert antibacterial effects, thereby achieving the goal of synergistic sterilization. |
