      目的 研究金银花、葛根、百合、薏苡仁、菊苣、姜黄等6种药食同源组合物的降尿酸作用。方法 利用中药系统药理学分析数据库、草药成分和靶标数据库等构建活性成分靶点网络图; 建立高尿酸小鼠模型, 检测小鼠血清中尿酸(uric acid, UA)、黄嘌呤氧化酶(xanthine oxidase, XOD)、腺苷脱氨酶(adenosine deaminase, ADA)、血尿素氮(blood urea nitrogen, BUN)、血清肌酐(creatinine, Cr)、肝脏XOD等生化指标。结果 根据网络药理学分析得出复方中包括44种活性成分, 其中核心靶点为ALB、AKT1和TNF, 其关键通路主要集中于糖脂代谢、炎症相关通路。动物实验结果表明, 与模型对照组(HUA)相比, 阳性对照组(API)、复方中剂量组(FM)、复方高剂量组(FH)的血清UA显著降低(P<0.05), 与HUA组相比, API组、复方低剂量组(FL)、FM组、FH组的血清ADA均显著降低(P<0.05)。在肾脏指标中, 与HUA组相比, API组、FL组、FM组、FH组的Cr水平显著降低(P<0.05)。结论 复方金银花组合物具有显著的降低UA以及肾保护的作用, 其作用机制可能与网络药理学提示的炎症通路、HIF-1信号通路、糖脂代谢及其相关通路和靶点调控有关。
英文摘要:
      Objective To investigate the uric acid lowering effects of 6 kinds of edible and medicinal resources including honeysuckle, pueraria, lily, coix seed, chicory and turmeric. Methods The target network map of active ingredients was constructed by searching the database of pharmacology analysis of database of pharmacology analysis of traditional Chinese medicine systems, herbal ingredients and target. Serum uric acid (UA), xanthine oxidase (XOD), adenosine deaminase (ADA), blood urea nitrogen (BUN), serum creatinine (Cr), liver XOD and other biochemical parameters in serum of mice were determined. Results According to network pharmacological analysis, the compound contained 44 kinds of active components, among which the core targets were ALB, AKT1 and TNF, and the key pathways were mainly related to glucose and lipid metabolism and inflammation. The results of animal experiments showed that compared with model control group (HUA), serum UA in positive control group (API), medium dose group (FM) and high dose group (FH) was significantly decreased (P<0.05). The serum ADA in API group, low dose group (FL), FM and FH group was significantly decreased (P<0.05). In renal indexes, compared with HUA group, Cr level in API group, FL group, FM group and FH group was significantly decreased (P<0.05). Conclusion This study reveale that the compound may play a role in lowering UA through glucose and lipid metabolism regulation, inflammation inhibition, kidney protection and so on.
