目的 构建大鼠膝骨关节炎模型, 考察含姜黄素和益生菌的蛋白固体饮料对大鼠膝骨关节炎模型的影响。方法 采用碘乙酸钠诱导大鼠膝骨关节炎模型, 按照“骨动力”蛋白固体饮料低剂量2.08 g/(kg·d)和高剂量4.16 g/(kg·d)给予大鼠灌胃给药, 阴性对照物为生理盐水, 阳性对照药物为双氯芬酸钠缓释片, 以大鼠右后肢机械刺激痛阈值、红外刺激痛阈值及软骨病理组织学观察为指标, 评价受试物对大鼠骨关节炎的影响。结果 对碘乙酸钠致大鼠骨关节炎模型的影响: 与模型组比较, 蛋白固体饮料在2.08 g/(kg·d)、4.16 g/(kg·d)剂量下给药13 d均能显著升高大鼠红外刺激足底痛阈值和机械刺激足底痛阈值。病理结果显示: 与模型组比较, 蛋白固体饮料4.16 g/(kg·d)剂量组大鼠的软骨表面结构、软骨细胞数量、软骨基质染色及病理评分等均有明显改善。结论 在实验所设剂量及研究条件下, “骨动力”蛋白固体饮料对碘乙酸钠诱导大鼠骨关节炎具有一定的治疗作用, 其作用机制可能与肠-关节轴理论有关。 |

bjective To establish a rat knee osteoarthritis model and investigate the effects of a protein solid beverage containing curcumin and probiotics on the rat osteoarthritis model. Methods The rat knee osteoarthritis model was induced by sodium iodoacetate. Rats were orally administered low-dose [2.08 g/(kg·d)] or high-dose [4.16 g/(kg·d)] “bone power” protein solid beverage. Physiological saline served as the negative control, while sustained-release diclofenac sodium tablets were the positive control. The efficacy of the test substance was evaluated using indicators such as the mechanical and infrared pain thresholds of the right hind limb, and histopathological observations of the cartilage. Results Effects on sodium iodoacetate-induced knee osteoarthritis in rats: Compared with the model group, administration of the protein solid beverage at doses of 2.08 g/(kg·d) and 4.16 g/(kg·d) for 13 d significantly increased the infrared stimulation plantar pain threshold and mechanical stimulation plantar pain threshold in rats. Pathological results showed that, compared with the model group, the structure of the cartilage surface, the number of chondrocytes, the staining of the cartilage matrix, and the pathological score were all markedly improved in the 4.16 g/(kg·d) dose group of the protein solid beverage. Conclusion At the dosages and under the research conditions set in the experiment, the “bone power” protein solid beverage demonstrates therapeutic effects on sodium iodoacetate-induced knee osteoarthritis in rats, and its mechanism of action may be related to the gut-joint axis theory. |
