      目的 研究料液比、剪切转速、剪切时间、剪切次数对蕨麻多糖提取率的影响, 评估其最佳提取率条件。方法 以青海蕨麻为试材, 采用高速剪切辅助提取技术, 单因素结合Box-Behnken响应面法对蕨麻多糖的最佳提取工艺条件进行优化, 并对其抗氧化活进行评价, 测定蕨麻多糖的1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazine, DPPH)自由基清除率、羟基自由基清除率、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐[2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt, ABTS]阳离子自由基清除率及总抗氧化能力。结果 在料液比1:40 (g/mL), 剪切转速19000 r/min, 剪切时间50 s, 剪切次数3次条件下得到的蕨麻多糖提取率为(25.580.42)%, 理论提取率为26.87%, 差值较小, 说明提取工艺参数可靠。体外抗氧化活性表明, 在试验浓度范围内, 蕨麻多糖对DPPH自由基、羟自由基、ABTS阳离子自由基有良好的清除作用, 且清除效果与多糖浓度呈剂量-效应关系。结论 蕨麻多糖具有较好的抗氧化活性, 本研究为蕨麻的进一步开发和应用提供数据支持。

      Objective To investigate the effects that the material-liquid ratio, rotational speed, shear time, and the number of shears had on the extraction rate of Potentilla anserina L. polysaccharides, and subsequently evaluate the optimal conditions. Methods Using Qinghai Potentilla anserina L. as the test material, the optimal extraction process conditions for Potentilla anserina L. polysaccharides were optimized through high-speed shear-assisted extraction technology using a one-way combination of Box-Behnken response surface methodology. Subsequently, their antioxidant properties were evaluated, determining the capabilities of scavenging 1,1-diphenyl-2- picrylhydrazine (DPPH) radicals, hydroxyl radicals, and 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) anion radicals, as well as assessing the total antioxidant capacity. Results The extraction rate of Potentilla anserina L. polysaccharide, when subjected to the conditions of a material-liquid ratio of 1:40 (g/mL), a shear speed of 19000 r/min, a shear time of 50 s, and a shearing process repeated 3 times, was found to be (25.58±0.42)%. This was closely aligned with the theoretical extraction rate of 26.87%, exhibiting a negligible discrepancy, thereby indicating the reliability of the extraction process parameters. In vitro antioxidant activity assessments revealed that the Potentilla anserina L. polysaccharide effectively scavenged DPPH radicals, hydroxyl radicals, and ABTS anion radicals within the experimental concentration range. Furthermore, this scavenging effects demonstrated a dose-dependent relationship with the concentration of the polysaccharide. Conclusion Potentilla anserina L. polysaccharide has been conclusively shown to exhibit good antioxidant activity and this study provides data support for further development and application of Potentilla anserina L.
