目的 探究人参提取物与水溶性番茄浓缩物复合剂对高脂饲料诱导大鼠的降血脂及抗氧化功效。方法 将40只Sprague-Dawley (SD)大鼠按照体重随机分为空白对照(blank control, CON)组和模型组,CON组提供维持饲料,模型组提供高脂饲料,持续14 d。然后将模型组随机分为模型对照(model,MOD)组、阳性对照(positive control,PC)组、人参提取物(ginseng extrac,GIN)组和人参提取物+水溶性番茄浓缩物复合剂(ginseng extract+water-soluble tomato concentrate,GIN+TOM)组。PC组、GIN组和GIN+TOM组提供高脂饲料,每日一次灌胃给药。灌胃45 d后测定大鼠体重、血清总胆固醇(total cholesterol, TC)、甘油三酯(total glyceride, TG)、高密度脂蛋白胆固醇(high density liptein cholesterol, HDL-C)、低密度脂蛋白胆固醇(low density lipoprotein, LDL-C)、低密度脂蛋白受体(low density lipoprotein receptor, LDLR)、羟甲基戊二酸单酰辅酶A还原酶(3-hydroxy-3-methyl glutaryl coenzyme A reductase, HMG-COAR)、胆固醇7α-羟化酶(cholesterol 7α-hydroxylase, CYP7A1)、丙二醛(malondialdehyde, MDA)等。结果 各组之间体重均无显著性差异(P>0.05)。MOD组与CON组相比,血清TC、TG、LDL-C和动脉粥样硬化指数水平均显著升高,HDL-C、超氧化物歧化酶(superoxide dismutase, SOD)、谷胱甘肽(glutathione, GSH)和谷胱甘肽过氧化物酶(glutathione peroxidase, GSH-PX)水平均显著降低。与MOD组相比,PC组、GIN组和GIN+TOM组血清TC、TG、LDL-C、MDA、HMG-COAR和动脉粥样硬化指数水平均显著降低;SOD、CYP7A1和LDLR水平显著升高。结论 人参提取物和水溶性番茄浓缩物复合剂均能够显著改善高脂饲料诱导的大鼠血脂升高等问题,其机制与降低血清HMG-COAR水平、抑制胆固醇合成及提高血清CYPA71水平、促进胆固醇降解有关,GIN+TOM组的效果优于GIN组。 |

Objective To investigate the effects of ginseng extract and water-soluble tomato concentrate on hyperlipidemia and antioxidation in rats induced by high fat diet. Methods Forty Sprague Dawley (SD) rats were randomly divided into a blank control (CON) group and a model group based on body weight. The CON group received maintenance feed, while the model group received high-fat feed for 14 days. Then, the model group was randomly divided into a model control (MOD) group, a positive control (PC) group, a ginseng extract (GIN) group, and a ginseng extract+water-soluble tomato concentration (GIN+TOM) group. The PC group, GIN group, and GIN+TOM group were provided with high-fat feed and administered orally once a day. After 45 days of gavage, the body weight, serum total cholesterol (TC), total triglycerides (TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), low density lipoprotein receptor (LDLR), hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-COAR), cholesterol 7α-hydroxylase (CYP7A1), and malondialdehyde (MDA), etc were measured. Results There was no significant difference in weight between the groups (P>0.05). Compared with CON group, the levels of serum TC, TG, LDL-C and atherosclerosis index in MOD group were significantly higher, while the levels of HDL-C, superoxide dismutase (SOD), glutathione (GSH) and glutathione peroxidase (GSH-PX) were significantly lower. Compared with MOD group, the levels of serum TC, TG, LDL-C, MDA, HMG-COAR and atherosclerosis index in PC group, GIN group and GIN+TOM group were significantly lower; the levels of SOD, CYP7A1, and LDLR significantly increased. Conclusion The combination of ginseng extract and water-soluble tomato concentrate can significantly improve the problem of high blood lipid elevation induced by high-fat diet in rats. The mechanism is related to reducing serum HMG-COAR level, inhibiting cholesterol synthesis, increasing serum CYPA71 level and promoting cholesterol degradation. The effect of GIN+TOM group is better than that of GIN group. |
