目的 研究海南地区大米中硒含量和存在形态情况。方法 利用电感耦合等离子体串联质谱仪,采用氧气反应模式(O2模式),测定海南地区大米的总硒含量;利用高效液相色谱-电感耦合等离子体串联质谱仪,采用氧气反应模式,测定海南地区大米中硒形态含量。结果 相较于常规的电感耦合等离子体质谱仪,电感耦合等离子体串联质谱仪在O2模式下,以最高丰度同位素80Se为检测目标质量数,其检测大米中硒的含量更加准确。228批次海南地区大米硒含量平均值为(0.0484±0.0728) mg/kg,约42%的大米达到GB/T 22499—2008《富硒稻谷》规定的富硒大米标准。随机对50批海南富硒大米的硒形态进行分析,检出的硒形态主要为硒代蛋氨酸(SeMet),少量大米检出硒代胱氨酸,其中硒代蛋氨酸(以Se计)的含量范围为0.0024~0.0255 mg/kg,占总硒含量的百分比为1.7%~51.4%。结论 海南地区大米具有一定的硒含量基础,部分大米硒代蛋氨酸含量占比较高,可开发成优质的富硒大米。 |

Objective To study the content and speciation of selennium in rice in Hainan area. Methods The total selenium content of rice in Hainan area was determined by inductively coupled plasma tandem mass spectrometry (ICP-MS/MS) in oxygen reaction mode (O2 mode). Using high performance liquid chromatography-inductively coupled plasma tandem mass spectrometry (HPLC-ICP-MS/MS) in oxygen reaction mode, the selenium speciation in rice in Hainan area was determined. Results Compared with conventional inductively coupled plasma mass spectrometry, ICP-MS/MS in O2 mode, with the highest abundance isotope 80Se as the detection target mass number, was more accurate in detecting the selenium content in rice. The average selenium content in 228 batches of rice in Hainan is (0.0484±0.0728) mg/kg, and about 42% of the rice meets the Se-enriched rice standard specified in the Selenium-enriched rice (GB/T 22499-2008). The selenium speciation of 50 batches of Hainan Se-enriched rice were randomly analyzed. The selenium speciation detected were mainly selenomethionine (SeMet), and selenocystine was detected in a small amount of rice. The content of SeMet (calculated as Se) ranged from 0.0024 to 0.0255 mg/kg, accounting for 1.7% to 51.4% of the total selenium content. Conclusion The rice in Hainan has a certain selenium content basis, and some of the rice has a high content of SeMet, which can be developed into high-quality Se-enriched rice. |
