目的:建立一种用高效液相色谱同时检测水果加工品中常见防腐剂、甜味剂和合成着色剂含量的分析方法。方法:称取匀质样品,调节pH值,混合溶剂提取后高速离心,氮吹、定容过滤待测。以甲醇、10mmol/L乙酸铵为主要流动相梯度洗脱,经Agilent ZORBAX Eclipse Plus C18色谱柱分离, 二极管阵列检测器多波长检测,外标法定量。结果:18种目标物在24min内均获得较好分离,0.2μg/mL~50μg/mL 的浓度范围内线性相关系数均在 0.9999 以上,方法检出限为0.0625mg/kg~2mg/kg,定量限为0.1875mg/kg~6mg/kg。在低、中、高3个水平,样品加标回收率为80.0%~119.9%, 相对标准偏差低于5.0%。结论:该方法适用基质种类较多,前处理简单易行,灵敏度和重复性较好,能准确测定水果加工品中相关食品添加剂的含量,对大量样品中常见食品添加剂的快速筛选具有实用价值。 |

Objective:Establish an analytical method for the simultaneous detection of common preservatives, sweeteners, and synthetic colorants in processed fruits using high-performance liquid chromatography. Methods:Weigh the homogenate sample, adjust the pH value, mix Liquid extraction and then centrifugate at high speed, nitrogen blow and constant volume filtration to be tested. Methanol and 10mmol/L Ammonium acetate were used as the main mobile phase for gradient elution, separated by Agilent ZORBAX Eclipse Plus C18 chromatographic column, detected by DAD at multi wavelength, and quantified by external standard method. Results:18 target substances were well separated within 24 minutes. The linear correlation coefficients within the concentration range of 0.2μg/mL~50μg/mL were all above 0.9999. The detection limit was 0.0625mg/kg~2mg/kg, and the quantitative limit was 0.1875mg/kg~6 mg/kg. At the three levels of low, medium, and high, the recovery rate of sample spiking is 80.0%~119.9%, and the relative standard deviation is less than 5.0%. Conclusion:The method is suitable for a wide variety of substrates, with simple and easy pre-treatment, good sensitivity and repeatability. It can accurately determine the content of related food additives in fruit processed products, and has practical value for rapid screening of common food additives in a large number of samples. |
