      目的 针对现有咪鲜胺快检技术不适用山药的问题,建立山药中咪鲜胺残留的快速检测方法,并应用于实际样品检测。方法 优化前处理技术,将胶体金快速检测方法应用于山药基质,验证方法的检出限、特异性、假阴性率和假阳性率,并通过液相色谱-串联质谱法确认方法的准确性。结果 建立的胶体金快速检测方法对山药中检出限为0.3 mg/kg,假阳性率2%,假阴性率0;与咪鲜胺-脱氨基咪唑、咪鲜胺锰盐和咪鲜胺-脱咪唑甲酰胺基的交叉反应率分别为200%、100%、100%,可同时检测咪鲜胺及其含有2,4,6-三氯苯酚部分的代谢物;对实际样本的检测结果与液相色谱-串联质谱法的符合率为88.2%。结论 本方法操作简便、检测速度快、准确性高,可应用于基层行政、农产品交易市场或餐饮配送公司快检实验室,从源头上保障食品安全。

      ABSTRACT:Objective? To solve the problem that the existing rapid detection technology of prochloraz is not suitable for Chinese yam, a rapid detection method of prochloraz residue in Chinese yam was established and applied to the detection of actual samples. Methods? By optimizing the pretreatment technique, the colloidal gold rapid detection method was applied to the Chinese yam matrix. The detection limit, specificity, false negative rate and false positive rate of the method were verified, and the accuracy was confirmed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Results? The detection limit of prochloraz in Chinese yam was 0.3 mg/kg, with the false positive rate 2% and the false negative rate 0. The cross-reaction rates with prochloraz-desimidazole-amino, prochloraz-manganese chloride complex and prochloraz- desimidazole-formylamino were 200%, 100% and 100%, respectively, and could simultaneously detect prochloraz and its metabolites containing 2,4,6-trichlorophenol. The coincidence rate between the test results of actual samples and LC-MS/MS was 88.2%. Conclusion? The method has the advantages of simple operation, fast detection speed and high accuracy, and can be applied to the fast inspection laboratory in grass-roots administration, agricultural products trading market or catering distribution companies to ensure food safety from the source.
