目的 建立有效测定山楂叶类保健食品中总黄酮含量的方法, 同时研究山楂叶总黄酮的抗氧化功效。方法 以胶囊剂为例, 研究山楂叶总黄酮提取的料液比例、溶剂乙醇体积分数和水浴蒸发温度, 并采用响应面分析法优化最佳提取条件。验证方法的线性、重复性、精密度、稳定性、准确度。对抗氧化性进行研究, 测定山楂叶总黄酮的1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazine, DPPH)自由基清除率、羟基自由基清除率、超氧阴离子自由基清除率。结果 建立以料液比0.025 (g/mL)、溶剂乙醇体积分数65%、水浴温度65℃的山楂叶总黄酮提取处理方法。建立的方法总黄酮重复性良好, 平均含量为1.3 g/100 g, 相对标准偏差(relative standard deviation, RSD)为2.6%, 仪器精密度为0.02% (n=6)。实验方法总黄酮回收率在90.57%~94.18%之间, RSD为1.3%。山楂叶总黄酮DPPH自由基、羟基自由基、超氧阴离子自由基清除效率分别能达到88.92%、84.46%、70.95%。结论 本研究建立的山楂叶总黄酮检测方法结果准确, 得出山楂叶总黄酮具有较强的抗氧化功效, 为高效快速测定以山楂叶为原料的保健食品中总黄酮的含量提供理论依据。 |

Objective To establish an effective method to determine the content of total flavonoids in hawthorn leaf health food, and study the antioxidant effects of total flavonoids in hawthorn leaf. Methods Taking capsule formulation as an example, the extraction ratio of total flavonoids from hawthorn leaves, solvent ethanol volume fraction, and water bath evaporation temperature were studied, and the optimal extraction conditions were optimized using response surface analysis. Verify the linearity, repeatability, precision, stability, and accuracy of the verification method. Study on antioxidant properties and determine the free radical scavenging rate of 1,1-diphenyl- 2-picrylhydrazine (DPPH), hydroxyl radical scavenging rate, and superoxide anion radical scavenging rate of total flavonoids in hawthorn leaves. Results A method for extracting total flavonoids from hawthorn leaves was established using a solid-liquid ratio of 0.025 (g/mL), a solvent ethanol volume fraction of 65%, and a water bath temperature of 65℃. The established method had good repeatability in total flavonoids, with an average content of 1.3 g/100 g, a relative standard deviation (RSD) of 2.6%, and an instrument precision of 0.02% (n=6). The total flavonoid recovery rate of the experimental method ranged from 90.57% to 94.18%, with an RSD of 1.3%. The scavenging efficiency of total flavonoids in hawthorn leaves against DPPH free radicals, hydroxyl free radicals, and superoxide anion free radicals could reach 88.92%, 84.46%, and 70.95%, respectively. Conclusion The detection method for total flavonoids in hawthorn leaves established in this study is accurate, and it is found that total flavonoids in hawthorn leaves have strong antioxidant effects, providing a theoretical basis for efficient and rapid determination of the content of total flavonoids in health foods made from hawthorn leaves. |
