      目的 分离纯化滁菊多糖(Chuzhou chrysanthemum polysaccharides, CCPS), 对其进行结构解析, 并研究其免疫活性。方法 以滁菊为原料, 热水浸提, 乙醇沉淀得到CCPS; 通过快流速二乙氨基乙基-琼脂糖凝胶(diethylaminoethyl-sepharose fast flow, DEAE-FF)和葡聚糖凝胶G-100 (Sephadex G-100)层析柱分离纯化CCPS, 得到中性多糖CCPS-1-1。用高效液相色谱法、紫外和红外光谱法、刚果红实验等进行CCPS-1-1结构解析, 建立小鼠单核巨噬细胞(mouse mononuclear macrophages cells, RAW264.7)的免疫模型探究其免疫刺激活性。结果 CCPS-1-1主要由鼠李糖、阿拉伯糖、半乳糖、半乳糖醛酸组成, 具有三股螺旋结构。体外免疫活性实验表明, CCPS-1-1可以促进RAW264.7细胞的增殖和吞噬作用, 显著诱导RAW264.7细胞中一氧化氮(nitric oxide, NO)的分泌及细胞因子包括肿瘤坏死因子-α (tumor necrosis factor-α, TNF-α)、白细胞介素-1β (interleukin-1β, IL-1β)和白细胞介素-6 (interleukin-6, IL-6)的释放。结论 分离纯化得到的滁菊CCPS-1-1具有促进免疫活性的作用, 在调节机体免疫力方面具有一定的应用价值。

      Objective To isolate and purify Chuzhou chrysanthemum polysaccharides (CCPS), analyze its structure, and study its immune activity. Methods Using Chuzhou chrysanthemum as raw material, CCPS was obtained by hot water extraction and ethanol precipitation. CCPS was isolated and purified by diethylaminoethyl- sepharose fast flow (DEAE-FF) and Sephadex G-100 chromatographic gel column, and CCPS-1-1 was obtained. High performance liquid chromatography, ultraviolet and infrared spectroscopy, and Congo red assays were used for structural elucidation of CCPS-1-1, and an immune model of mouse mononuclear macrophages cells (RAW264.7) was established to explore its immunostimulatory activity. Results CCPS-1-1 was mainly composed of rhamnose, arabinose, galactose, galacturonic acid, with a three-stranded helical structure. In vitro immunoreactivity assay showed that CCPS-1-1 promoted the proliferation and phagocytosis of RAW264.7 cells, and significantly induced the secretion of nitric oxide (NO) and the release of cytokines including tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) release in RAW264.7 cells. Conclusion The isolated and purified Chuzhou chrysanthemum CCPS-1-1 has the effect of promoting immunological activity, which has certain application value in regulating the immunity of the organism.
