      目的 通过比较不同植物试剂盒在花椒果皮DNA提取中的差异, 探究适用于花椒果皮的基于离心柱法的DNA提取试剂盒。方法 选取8种不同品种的花椒, 利用TIANGEN DNAsecure新型植物基因组DNA提取试剂盒(Tiangen-Ⅰ)、DNeasy Plant Mini kit (DNeasy)、多糖多酚植物基因组DNA提取试剂盒(Tiangen-Ⅱ)、Nucleospin ? Food kit (Nucleospin) 4种基于离心柱法的试剂盒提取花椒果皮基因组DNA, 通过对比提取到的DNA浓度、纯度、聚合酶链式反应扩增能力, 以及提取过程中的成本、耗时等因素, 基于简单加权(simple additive weighting, SAW)分析对比不同试剂盒对于花椒果皮DNA的提取效果, 并进行综合评分。结果 不同试剂盒各有优缺点, 最终Tiangen-Ⅰ试剂盒得分1.64, 排名第一, 综合考虑价格、所用时间与提取得到的DNA质量, Tiangen-Ⅰ试剂盒是更适用于花椒果皮DNA提取的试剂盒; 同时, Nucleospin试剂盒在提取得到的花椒DNA质量方面有着较好的表现。结论 本研究以花椒为例探究干扰组分较多植物的基因组DNA试剂盒提取方案, 通过比较Tiangen-Ⅰ试剂盒被推荐作为最佳选择, 本研究为花椒果皮及其他相似植物的基因组DNA提取提供了参考方案。

      Objective To explore the DNA extraction kit based on centrifugal column method suitable for Zanthoxylum pericarp, by comparing the differences of different plant kits in DNA extraction from Zanthoxylum pericarp. Methods Eight different varieties of Zanthoxylum were selected, and the genomic DNA of Zanthoxylum pericarp was extracted by four kinds of kits based on centrifugal column method, including TIANGEN DNAsecure new plant genomic DNA extraction kit (Tiangen-I), DNeasy Plant Mini kit (DNeasy), polysaccharide polyphenol plant genomic DNA extraction kit (Tiangen-II) and Nucleospin ? Food kit (Nucleospin). By comparing the extracted DNA concentration, purity, polymerase chain reaction amplification ability, as well as the cost and time-consuming factors in the extraction process, the effects of different kits on the extraction of Zanthoxylum pericarp DNA were analyzed and compared based on simple additive weighting (SAW). Results Different kits had their own advantages and disadvantages. Finally, Tiangen-I kit scored 1.64, ranking first. Considering the price, time and DNA quality, Tiangen-I kit was more suitable for DNA extraction from Zanthoxylum pericarp. At the same time, the Nucleospin kit had a good performance in the quality of the extracted Zanthoxylum DNA. Conclusion In this study, Zanthoxylum is used as an example to explore the genomic DNA kit extraction scheme of plants with more interference components. By comparing the Tiangen-I kit is recommended as the best choice, this study provide a reference scheme for the genomic DNA extraction of Zanthoxylum peel and other similar plants.
