      目的 探究果胶交联β-环糊精(pectin crosslinked β-cyclodextri, PT@β-CD)负载岩藻黄质(fucoxanthin, FC)在体外的降脂活性。方法 实验室前期制备的PT@β-CD负载FC (FC-PT@β-CD)通过扫描电子显微镜(scanning electron microscopy, SEM)、粒径分析(particle size analysis, PSA)、红外光谱(infrared spectroscopy, FT-IR)检测、热重分析(thermogravimetric analysis, TGA)进行表征, 并研究FC-PT@β-CD在体外对甘胆酸钠和牛磺胆酸钠的吸附以及对胰脂肪酶的抑制。结果 负载实验结果表明PT@β-CD/C的包封率和装载量为86.00%±0.71%和9.82%±0.12%。SEM结果显示FC-PT@β-CD呈球状, FC-PT@β-CD/A、FC-PT@β-CD/B、FC-PT@β-CD/C的平均粒径分别为248.4、199.0、157.0 nm, FT-IR结果证明FC成功负载在PT@β-CD, TGA结果表明FC-PT@β-CD/C在259.4℃下明显减重, 热稳定良好且体外释放稳定。FC-PT@β-CD对甘胆酸钠吸附率为25.25%±0.82%, 对牛磺胆酸钠吸附率为56.21%±1.04%。对胰脂肪酶的抑制率随着FC-PT@β-CD浓度增加而增加, 最后趋于平稳, 且为混合型抑制, 半抑制浓度(half maximal inhibitory concentration, IC50)为28.45 mg/mL。结论 FC-PT@β-CD在体外具有良好的降脂作用, 为岩藻黄质在食品中的应用和进一步开发提供了思路。

      Objective To investigate the in vitro hypolipidemic activity of pectin crosslinked β-cyclodextrin (PT@β-CD) loaded fucoxanthin (FC). Methods PT@β-CD loaded FC (FC-PT@β-CD) pre prepared in the laboratory was characterized by scanning electron microscopy (SEM), particle size analysis (PSA), infrared spectroscopy (FT-IR) detection and thermogravimetric analysis (TGA). The adsorption of sodium cholate and sodium taurocholate of FC-PT@β-CD and the inhibitory effect on pancreatic lipase were studied in vitro. Results The loading test showed that the encapsulation efficiencies and loading capacities of PT@β-CD/C were 86.00%±0.71% and 9.82%±0.12%. SEM results showed that FC-PT@β-CD was spherical. The average particle sizes of FC-PT@β-CD/A, FC-PT@β-CD/B and FC-PT@β-CD/C were 248.4, 199.0 and 157.0 nm, respectively. FT-IR results showed that FC was successfully loaded on PT@β-CD. TGA results showed that the weight of FC-PT@β-CD/C was significantly reduced at 259.4℃, thermal stability was good and in vitro release was stable. The adsorption rate of sodium cholate by FC-PT@β-CD was 25.25%±0.82%. The adsorption rate of sodium taurocholate was 56.21%±1.04%. The inhibition rate of pancreatic lipase increased with the concentration of FC-PT@β-CD, and finally reached a plateau. The inhibition was a mixed type, and the half maximal inhibitory concentration (IC50) was 28.45 mg/mL. Conclusion FC-PT@β-CD CD has a good hypolipidemic activity in vitro, providing ideas for the application and further development of fucoxanthin in food.
