目的 建立时间分辨荧光免疫分析法同时快速检测禽蛋中氟苯尼考、氟苯尼考胺的分析方法。方法 采用乙腈对禽蛋中氟苯尼考和氟苯尼考胺进行提取, 并用二氯乙酸酐将提取液中的氟苯尼考胺衍生化为氟苯尼考, 最后用时间分辨荧光免疫分析技术对氟苯尼考总量进行快速检测。结果 衍生试剂二氯乙酸酐体积分数为3%时, 氟苯尼考胺生成氟苯尼考含量最高, 副产物最少。氟苯尼考及氟苯尼考胺检出限为10 μg/kg。灵敏度、特异性和相对准确度为100%, 假阴性率和假阳性率均为0%。另外, 采用本研究建立的时间分辨荧光免疫分析技术对市售禽蛋样品进行检测, 其结果与食品安全国家标准检测结果基本一致。结论 本方法前处理简单、快速, 检测成本低、结果准确可靠, 适合基层日常监管, 重大活动食品安全的现场快速筛查、快速检测, 提高检测效率。 |

Objective To establish a time-resolved fluorescent immuno-chromatography for the simultaneous and rapid detection of florfenicol and florfenicol-amine in poultry eggs. Methods Florfenicol and florfenicol-amine were extracted from poultry eggs with acetonitrile, and florfenicol-amine was derived into florfenicol by dichloroacetic anhydride. Finally, florfenicol was detected by a time-resolved fluorescent immuno-chromatography card test strip. Results Under the condition of when the volume fraction of the derivative reagent dichloroacetic anhydride was 3%, the content of flufenicol derived from flufenicol amine was the highest, and the byproducts were the least, and the limits of detection of florfenicol and florfenicol-amine were 10 μg/kg, the sensitivity, specificity, and relative accuracy were 100%, and the false negative rate and false positive rate were all 0%. In addition, the time-resolved fluorescent immuno-chromatography established in this study was used to detect the eggs in the market, and the results were basically consistent with the national food safety standards. Conclusion This method has the advantages of simple and rapid pretreatment, low detection cost, accurate and reliable results, and is suitable for daily supervision at the grass-roots level, rapid screening and rapid detection of food safety at major events, and improves detection efficiency. |
