目的 优化加压辅助碱法提取蝉花子实体(人工培植)多糖的工艺, 并研究其免疫活性。方法 以多糖提取率为指标, 在单因素基础上, 经正交实验优化其提取工艺。同时以小鼠体重增长率、脏器指数, 血清中肿瘤坏死因子-α (tumor necrosis factor-α, TNF-α)、白细胞介素-2 (interleukin-2, IL-2)、白细胞介素-6 (interleukin-6, IL-6)的含量、免疫球蛋白A (immunoglobulin A, IgA)、免疫球蛋白G (immunoglobulin G, IgG)、免疫球蛋白M (immunoglobulin M, IgM)的含量以及小鼠脾淋巴细胞增殖刺激指数作为指标来评价免疫活性。结果 最佳提取工艺条件为: 氢氧化钠浓度0.2 mol/L、提取压力1.0 MPa、提取温度80℃、提取时间40 min, 在此条件下多糖提取率为8.68%, 该方法提取效果优于单一提取法。体外免疫活性实验表明, 蝉花子实体多糖能有效增加模型小鼠的体重增长率及脏器指数, 细胞因子(TNF-α、IL-2和IL-6)、免疫球蛋白(IgA、IgG和IgM)的含量和脾淋巴细胞增殖刺激指数, 表明其具有明显的提高免疫力作用。结论 优化后的提取条件能够有效提取蝉花子实体多糖, 所得多糖具有明显的提高免疫力作用。该研究可为蝉花子实体多糖的提取及开发应用提供科学依据。 |

Objective To optimize the process of Cordyceps cicadae fruiting body polysaccharides (artifical cultivation) by pressurization-assisted alkaline extraction method, and study its immunoactivity. Methods On the basis of single factor, the extraction process was optimized by orthogonal experiment with the extraction rate of polysaccharide as index. At the same time, the body weight growth rate, organ index of mice, and the levels of tumor necrosis factor-α (TNF-α), interleukin-2 (IL-2) and interleukin-6 (IL-6), the content of immunoglobulin immunoglobulin A (IgA), immunoglobulin G (IgG), immunoglobulin M (IgM) in serum, and mouse spleen lymphocyte proliferation stimulation index were used to evaluate immune activity. Results The optimal extraction conditions were as follows: Sodium hydroxide concentration was 0.2 mol/L, extraction pressure was 1.0 MPa, extraction temperature was 80℃, extraction time was 40 min. Under these conditions, the polysaccharides extraction rate was 8.68%, and the extraction effect of this method was better than that of the single extraction method. In vitro immunoactivity tests had shown that Cordyceps cicadae fruiting body polysaccharides could effectively increase the body weight growth rate and organ index of model mice, as well as the content of cytokine (TNF-α, IL-2 and IL-6), immunoglobulins (IgA, IgG and IgM), and the spleen lymphocyte proliferation stimulation index, which indicated that it had a significant immune enhancing effect. Conclusion The optimized extraction conditions can effectively extract Cordyceps cicadae fruiting body polysaccharides, and the obtained polysaccharides from Cordyceps cicadae fruiting body have a significant immune enhancing effect. This study can provide scientific basis for the extraction, development, and application of Cordyceps cicadae fruiting body polysaccharides. |
