目的 分析红芽木的化学成分, 并对其抗氧化、降糖及降脂功能活性进行研究。方法 采用液相色谱-质谱法采集数据, 对各色谱峰的质谱数据进行解析、结构推测和确认; 采用自由基清除能力方法对红芽木样品抗氧化活性进行研究; 考察红芽木对α-葡萄糖苷酶抑制活性及对胰岛素抵抗的影响; 通过建立人肝癌高脂模型测试红芽木的降脂活性。结果 从红芽木中共鉴定了68个化合物, 抗氧化结果显示红芽木清除1,1-二苯基-2-三硝基苯肼自由基及2,2’-联氮-二(3-乙基-苯并噻唑啉-6-磺酸)二铵盐阳离子自由基的半抑制浓度分别为(0.21±0.05) mg/mL、(0.14±0.03) mg/mL; 红芽木对α-葡萄糖苷酶抑制活性的半抑制浓度为(63.52±3.12) μg/mL; 降糖实验结果显示可以改善HepG2细胞的胰岛素抵抗; 降脂实验结果显示红芽木提取物具有一定的降脂作用。结论 红芽木化学成分丰富, 并具有较好的抗氧化、降糖及降脂活性, 本研究为红芽木的深入研究开发提供了参考。 |

Objective To analyze the chemical constituents of Cratoxylum formosum and study its antioxidant, hypoglycemic, and lipid-lowering activities. Methods Data were collected by liquid chromatography-mass spectrometry, and the mass spectrometry data of each chromatographic peak were analyzed, inferred and confirmed. The antioxidant activity of Cratoxylum formosum was studied by radical scavenging ability method. The inhibitory activity of Cratoxylum formosum on α-glucosidase and its effect on insulin resistance were investigated; the lipid-lowering activity of Cratoxylum formosum was tested by establishing a high-fat model of human liver cancer. Results A total of 68 compounds were identified from Cratoxylum formosum. The antioxidant results showed that the half maximal inhibitory concentrations of Cratoxylum formosum for scavenging 1,1-diphenyl-2-picrylhydrazyl radicals and 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) ammonium salt anion radicals were (0.21±0.05) mg/mL and (0.14±0.03) mg/mL, respectively. The semi-inhibitory concentration for α-glucosidase activity of Cratoxylum formosum was (63.52±3.12) μg/mL. The results of hypoglycemic experiment showed that it could improve the insulin resistance of HepG2 cells. The results of lipid-lowering experiment showed that the extract of Cratoxylum formosum had a certain lipid-lowering effect. Conclusion Cratoxylum formosum is rich in chemical components and has good antioxidant, hypoglycemic and lipid-lowering activities. This study provides a reference for the in-depth research and development of Cratoxylum formosum. |
