      目的 探究副干酪乳杆菌X12 (Lactobacillus. paracasei subsp. paracasei X12)抑制N-二甲基亚硝胺(N-nltrosodimethylamine, NDMA)诱导IEC-6细胞(大鼠肠道黏膜细胞)毒性损伤作用。方法 通过胃液、肠液耐受性实验、扫描电镜法分析副干酪乳杆菌X12益生功能和菌株的形态; 通过CCK-8实验对副干酪乳杆菌X12抑制NDMA诱导IEC-6细胞毒性作用进行分析。结果 胃肠道耐受性实验显示, 副干酪乳杆菌X12可耐受胃肠液的消化, 但对胆盐的耐受性略差; 扫描电镜下副干酪乳杆菌X12菌体呈短而圆的杆状结构, 无鞭毛; CCK-8实验结果显示, 在MRS培养基中NDMA (0~80 μg/mL)对副干酪乳杆菌X12的生长无显著影响(P>0.05); 副干酪乳杆菌X12可抑制NDMA致大鼠肠道黏膜细胞IEC-6毒性的作用, 且该作用与副干酪乳杆菌X12的剂量有关。结论 副干酪乳杆菌X12可抑制NDMA诱发的IEC-6细胞毒性损伤。

      Objective To explore the inhibitory effects of Lactobacillus. paracasei subsp. paracasei X12 on the toxic injury of IEC-6 cells (rat intestinal mucosal cells) induced by N-dimethylnitrosamine (NDMA). Methods The probiotic function and morphology of Lactobacillus. paracasei subsp. paracasei X12 were analyzed by gastric fluid and intestinal fluid tolerance test and scanning electron microscopy. The toxic effect of the strain X12 on NDMA-induced IEC-6 cells were analyzed by CCK-8 assay. Results Gastrointestinal tolerance test showed that Lactobacillus. paracasei subsp. paracasei X12 could tolerate the digestion of gastroenteric fluid, but had a slightly poor tolerance to bile salts. Under scanning electron microscope, the cell of Lactobacillus. paracasei subsp. paracasei X12 showed short and round rod-like structure without flagella. The results of CCK-8 experiment showed that NDMA (0?80 μg/mL) had no significant effect on the growth of the strain X12 in MRS medium. Lactobacillus. paracasei subsp. paracasei X12 inhibited the toxicity of IEC-6 cell of rat intestinal mucosal induced by NDMA, and the effect was related to the dose of Lactobacillus. paracasei subsp. paracasei X12. Conclusion Lactobacillus. paracasei subsp. paracasei X12 inhibited the cytotoxic damage of IEC-6 cells induced by NDMA.
