      目的 探究粉葛愈伤组织次生代谢产物的生物活性和功能成分。方法 分级制备石油醚提取物(petroleum ether extract, PE)、乙酸乙酯提取物(ethyl acetate extract, EE)、甲醇提取物(methanol extract, ME)和水提取物(water extract, WE), 采用体外抑菌法、自由基清除法和比色法评估其抗菌、抗氧化和细胞毒活性, 并分析总黄酮、总酚、总多糖及其单体等功能成分。结果 EE活性最佳, 对测试菌的最小抑菌浓度为0.195~1.563 mg/mL, 对1,1-二苯基-2-三硝基苯肼自由基和2,2’-联氮双(3-乙基苯并噻唑啉-6-磺酸)二铵盐阳离子自由基的半抑制浓度分别为0.2795 mg/mL和0.0640 mg/mL, 对人肺癌细胞A549的半最大效应浓度为260.9 μg/mL。总黄酮含量在EE中最高达到(273.89±7.48) μg/mL, 其中大豆苷和大豆苷元含量分别达到(30.3428±1.4251) μg/mL和(45.7235±2.0230) μg/mL。结论 EE具有较强的抗菌、抗氧化和微弱的细胞毒活性, 且其总黄酮含量高, 以大豆苷和大豆苷元为主, 该结果为粉葛愈伤组织在食品与制药领域的开发利用提供了一定的参考。

      Objective To explore the bioactivity and functional components of secondary metabolites from callus of Pueraria thomsonii. Methods The petroleum ether extract (PE), ethyl acetate extract (EE), methanol extract (ME) and water extract (WE) were prepared by grading, the antibacterial, antioxidant and cytotoxic activities were evaluated by in vitro bacteriostatic method, free radical scavenging method and colorimetric method, and functional components such as total flavonoids, total phenols, total polysaccharides and their monomers were analyzed. Results EE had the best activities, the minimum inhibitory concentration for the tested bacteria was 0.195?1.563 mg/mL, the half inhibitory concentration for 1,1-diphenyl-2-picrylhydrazyl radical and 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) ammonium salt cationic radical were 0.2795 mg/mL and 0.0640 mg/mL respectively, the concentration for 50% of maximal effect for human lung cancer cell A549 was 260.9 μg/mL. The highest content of total flavonoids in EE was (273.89±7.48) μg/mL, and the content of daidzin and daidzein were (30.3428±1.4251) μg/mL and (45.7235±2.0230) μg/mL, respectively. Conclusion EE have strong antibacterial, antioxidant and weak cytotoxic activities, and its total flavonoids content is high which mainly daidzin and daidzein. These results provide a certain reference for the development and utilization of callus of Pueraria thomsonii in food and pharmaceutical fields.
