重组酶介导等温核酸扩增(recombinase aided amplification, RAA)是一种新型的国产等温扩增技术, 其扩增反应可以在恒定温度(37~42℃)下快速完成, 具有较高检测效率, 且对精密仪器和检测场地的要求较低, 更能应对大批量筛查、应急处理等应用场景。经10年的科学研究和实践应用, RAA技术已取得了诸多突破性成果, 本文介绍了RAA技术的反应原理, 综述了近3年RAA技术及其扩展技术在食源性致病菌检测中的研究进展, 总结了RAA技术在样品前处理、假阳性和假阴性、检测通量、试剂盒开发等方面存在的技术难点, 并对未来的研究方向提出了若干建议, 以期为RAA技术在食源性致病菌快速检测中的应用推广提供研究思路。 |

Recombinase aided amplification (RAA) is a new type of domestic isothermal amplification technology. Its amplification reaction can be rapidly completed at a constant temperature (37?42℃), with high detection efficiency and low requirement on precision instruments and detection sites. More suitable for large-scale screening, emergency treatment and other application scenarios. Through 10 years of scientific research and practical application, RAA technology has made many breakthrough achievements. This paper introduced the reaction principle of RAA technology, and reviewed the research progress of RAA technology and its extension technology in the detection of food-borne pathogenic bacteria in the past three years. Besides, this paper proposed the technical difficulties of RAA technology in sample pretreatment, false positiveand false negative, detection flux, kit development, and put forward some suggestions for future research directions, in order to provide research ideas for the application and popularization of RAA technology in rapid detection of foodborne pathogens. |
