目的 研究蒲公英红枣汁对D-半乳糖致衰老模型雌性大鼠体内抗氧化、抗炎作用的影响。方法 50只雌性大鼠随机分为对照组、模型组、蒲公英红枣汁低(5 mL/kg)、中(10 mL/kg)、高(20 mL/kg)剂量组。对照组以外的大鼠腹腔注射D-半乳糖500 mg/(kg·d), 连续6周建立衰老模型。然后蒲公英红枣汁灌胃干预30 d, 对照组与模型组以同样方法给予等体积蒸馏水。检测各组大鼠血清和卵巢组织丙二醛(malondialdehyde, MDA)含量和超氧化物歧化酶(superoxide dismutase, SOD)、血清谷胱甘肽过氧化物酶(glutathione peroxidase, GSH-Px)、过氧化物酶(catalase, CAT)活性、谷胱甘肽(glutathione, GSH)含量等抗氧化相关指标; 测定大鼠血清中白细胞介素1β (interleukin-1β, IL-1β)、白细胞介素-6 (interleukin-6, IL-6)、肿瘤坏死因子(tumor necrosis factor-α, TNF-α)、诱导型一氧化氮合酶(inducible nitric oxide synthase, iNOS)含量及外周血T淋巴细胞亚群水平。评价蒲公英红枣汁体外抗氧化能力(oxygen radical absorbance capacity, ORAC)。结果 蒲公英红枣汁各组均能显著降低血清和卵巢组织MDA含量(P<0.05), 提高血清GSH含量、GSH-Px活性(P<0.01), 降低血清IL-1β水平(P<0.05), 降低CD4+/CD8+比值(P<0.05); 低、中剂量组降低了血清蛋白质羰基含量(P<0.05); 低、高剂量组提高了SOD活性(P<0.05); 高剂量组降低了TNF-α水平(P<0.01)。低、中、高剂量3组未显示出剂量依赖关系。结论 蒲公英红枣汁可以提高D-半乳糖衰老模型雌性大鼠的抗氧化和抗炎能力, 可能通过抑制体内氧化应激及提高机体抗炎因子水平发挥延缓衰老的作用。 |

Objective To study the effects of dandelion jujube juice on antioxidant and anti-inflammatory in female rats aging model induced by D-galactose. Methods Fifty female rats were randomly divided into the control group, model group, dandelion jujube juice low (5 mL/kg), medium (10 mL/kg), high (20 mL/kg) dose group. Rats other than the control group were intraperitoneally injected with D-galactose 500 mg/(kg·d) for 6 weeks to establish aging models. Then the dandelion jujube juice was administered by gavage for 30 days, the control group and the model group were given the same volume of distilled water. Malondialdehyde (MDA) content in serum and ovarian tissue and superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT) activity, and glutathione (GSH) content in serum and ovarian tissue of rats in each group were detected. The content interleukin-1β (IL-1β), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), inducible nitric oxide synthase (iNOS) and T lymphocyte subsets in peripheral blood of rats were measured. The oxygen radical absorbance capacity (ORAC) of dandelion jujube juice was measured in vitro. Results Dandelion jujube juice could significantly reduce the MDA content in serum and ovarian tissue (P<0.05), improve the serum GSH content and GSH-Px activity (P<0.01), reduce serum IL-1β level (P<0.05), and reduce the CD4+/CD8+ ratio (P<0.05). The serum protein carbonyl content was decreased in the low and medium dose groups (P<0.05). The SOD activity was increased in the low-dose and high-dose groups (P<0.05). The level of TNF-α was decreased in the high dose group (P<0.01). The low, medium and high dose groups did not show a dose-dependent relationship. Conclusion Dandelion jujube juice can improve the antioxidant and anti-inflammatory capacity of female rats with D-galactose aging model, which may play a role in delaying aging by inhibiting oxidative stress in vivo and increasing the level of anti-inflammatory factors in the body. |
