      目的 建立一种基于电化学参数分析的恒电位仪系统用于快速准确低成本地检测5’-鸟苷酸。方法 以Ru(bpy)22+/SMWCNTs/Au复合材料修饰的玻碳电极为媒介, 筛选出与电子发生还原反应的靛蓝作为显色剂, 研究了溶液pH和靛蓝添加量体积比对靛蓝电化学还原性能的影响, 经过条件优化后构建一种使靛蓝脱色生成隐色体靛白的双电极体系恒电位装置。结果 在最佳检测条件下, 当5’-鸟苷酸线性范围在0~0.6 mmol/L时, 靛蓝脱色率随5’-鸟苷酸浓度的增大而呈线性上升(r2=0.9614), 该方法在肉汤样品中5’-鸟苷酸的检测过程中, 显示出与电化学传感器技术高度相似的结果。结论 本方法能够有效地在实际应用过程中现场检测鲜味核苷酸, 因其操作简便、便携微型化, 在未来的食品质量检测领域将具有较大的市场优势与应用价值。
英文摘要:
      Objective To establish a potentiostat system based on electrochemical parameter analysis for rapid, accurate and low-cost detection of 5’-guanosine acid. Methods A glassy carbon electrode modified with Ru(bpy)22+/SMWCNTs/Au composite was used as a medium to select indigo which has a reduction reaction with electrons as a chromogenic agent. The effects of solution pH and volume ratio of indigo addition on the electrochemical reduction performance of indigo were investigated, and a two-electrode system constant potential device was constructed to decolorize indigo to produce the cryptophore indigo white after optimization of conditions. Results Under the optimal detection conditions, when the linear range of 5’-guanosine acid was from 0 to 0.6 mmol/L, the indigo decolorization rate increased linearly with increasing 5’-guanosine acid concentration (r2=0.9614), and the method showed highly similar results to electrochemical sensor technology for the detection of 5’-guanosine acid in broth samples. Conclusion This method can be effectively applied in the field of fresh nucleotide detection, which is easy to operate, portable and miniaturized, and will have a great market advantage and application value in the field of food quality testing in the future.
