目的 建立食品中典型酰胺醇类抗生素残留的快速检测方法并对该类药物在渝东北地区的残留污染风险进行评估。方法 采用乙腈萃取食品基质中残留的酰胺醇类抗生素, QuEChERS法进行净化, 超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)分析检测; 同时应用层次分析法(analytic hierarchy process, AHP)对渝东北地区食品中酰胺醇类药物残留的污染风险进行评估。结果 QuEChERS-UPLC-MS/MS检测技术精确度高, 氯霉素、氟苯尼考和甲砜霉素在0.15~50.00 μg/kg浓度范围内的线性相关系数均大于0.999, 在0.15~50.00 μg/kg浓度水平范围内的回收率分别为85.2%~100.9%、76.7%~90.6%、85.3%~96.7%, 精密度均小于15%, 检出限为0.10 μg/kg, 定量限为0.15 μg/kg。求解AHP模型可知, 渝东北地区酰胺醇类药物残留的食品污染风险程度为: 氟苯尼考>甲砜霉素>氯霉素。其中, 氟苯尼考: 畜副产品>鲜蛋>水产品>禽副产品>禽肉>畜肉; 甲砜霉素: 禽副产品>鲜蛋>禽肉; 氯霉素: 畜副产品>水产品>畜肉。结论 QuEChERS-UPLC-MS/MS适用于对食品中酰胺醇类抗生素残留的定性定量检测, AHP评价法则可对可能引发的污染风险大小进行综合评估, 相关检测方法和评价模型能为区域食品安全的风险管控提供支撑。 |

Objective To establish a rapid detection method for typical aminool antibiotic residues in food in northeast Chongqing and to evaluate the risk of such drug residues. Methods Acetonitrile solution was used to extract the residues of aminool antibiotics in the food matrix, and then the QuEChERS method was applied for extraction and purification, ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) analysis and detection. Additionally, the analytic hierarchy process (AHP) evaluation model aimed to assess safety risk of amide alcohol drug residues. Results The results showed that the detection technology of QuEChERS- UPLC-MS/MS was highly accurate, in which the linear correlation coefficients of chloramphenicol, florfenicol and thiamphenicol were all greater than 0.999 in the concentration ranging from 0.15 to 50.00 μg/kg. The recoveries were 85.2%?100.9%, 76.7%?90.6%, and 85.3%?96.7% in the range of 0.15 to 50.00 μg/kg respectively, the precision was less than 15%, meanwhile, the limit of detection was 0.10 μg/kg, with the limit of quantitation being 0.15 μg/kg. According to the results of the AHP evaluation model, the risk degree of food contamination of amide alcohol residues in northeast Chongqing was as follows: Florfenicol>thiamphenicol>chloramphenicol. Among them, florfenicol: Livestock byproducts>fresh eggs>aquatic products>poultry byproducts>poultry meat>livestock meat, thiamphenicol: Poultry byproducts>fresh eggs>poultry meat, and chloramphenicol: Livestock byproducts>aquatic products>livestock meat. Conclusion QuEChERS-UPLC-MS/MS is suitable for qualitative and quantitative detection of amido alcohol antibiotic residues in food, AHP can comprehensively evaluate the possible pollution risks, and the related detection methods and evaluation models can provide support for risk management and control of regional food safety. |
