目的 比较分析人工培育杂交河鲀(红鳍东方鲀♀×暗纹东方鲀♂)和养殖红鳍东方鲀的肌肉品质。方法 以杂交河鲀和红鳍东方鲀为实验原料, 分别测定肌肉质构特性、肌糖原含量、腺嘌呤核苷三磷酸(adenosine triphosphate, ATP)及其关联化合物水平、游离氨基酸和肌原纤维蛋白热稳定性指标, 通过数据分析进行综合评价。结果 杂交河鲀肌肉硬度(28.80 N)显著低于红鳍东方鲀(31.58 N)(P<0.05)。杂交河鲀肌肉的游离氨基酸组成以赖氨酸、丙氨酸和甘氨酸为主, 和红鳍东方鲀组成基本相似。此外, 红鳍东方鲀比杂交河鲀肌原纤维蛋白稳定性更好, 在45℃下钙ATP酶失活速率分别为3.3×10?3/s和6.1×10?3/s。胰凝乳蛋白酶对两种肌原纤维蛋白的酶解图谱基本相似, 肌球蛋白分别在低盐条件下被酶解为的S-1和rod片段或在高盐条件下被酶解为肌球蛋白重酶解链和轻酶解链。结论 杂交河鲀肌肉质地更柔软, 与红鳍东方鲀肌肉具有相似的氨基酸组成,两者的肌球蛋白结构相似, 都具有较好的热稳定性。 |

Objective To compare the muscle quality characteristics of artificial hybrid pufferfish (Takifugu rubripes ♀ × Takifugu obscurus ♂) with the cultured Takifugu rubripes. Methods Hybrid pufferfish and Takifugu rubripes were used as experiments materials. The indexes of texture analysis, glycogen content, adenosine triphosphate (ATP) and its related compounds, free amino acids, and thermal stability of myofibrillar protein were measured, respectively, and through data analysis conducted a comprehensive evaluation. Results The muscle hardness of hybrid pufferfish was 28.80 N, which was significantly lower than that of Takifugu rubripes, with the value of 31.58 N (P<0.05). The free amino acid composition of the hybrid pufferfish muscle was mainly lysine, alanine and glycine, which was basically similar to that of the Takifugu rubripes. In addition, the stability of Takifugu rubripes was better than that of hybrid pufferfish, and the deactivation rates of calcium ATPase at 45℃ were 3.3×10?3/s and 6.1×10?3/s, respectively. The enzymolysis patterns of the 2 kinds of chymotrypsin were basically similar. Myosin was enzymolized into S-1 and rod fragments under low salt conditions, or heavy and light myosin chains under high salt conditions. Conclusion The hybrid pufferfish has a softer muscle texture and has a similar free amino acid composition. and myosin structure comparing to Takifugu rubripes. Both of them have better thermal stability. |
