目的 比较分析两种雪菊的黄酮化合物成分和关键酶基因表达的差异。方法 依据超高效液相色谱-串联四极杆飞行时间质谱法对雪菊中的黄酮类代谢产物进行分析鉴定, 同时采用实时定量PCR法(quantitative real-time PCR, RT-qPCR)对黄酮合成关键酶基因进行表达差异分析。结果 从雪菊中共分析鉴定出204个黄酮类成分, 并筛选出15种差异表达的黄酮类代谢物, 其中6种黄酮在平原雪菊中含量较高, 9种黄酮在高原雪菊中含量较高。通过对差异代谢物的通路富集分析, 发现差异黄酮代谢物显著富集于类黄酮生物合成、次生代谢物的生物合成、异黄酮生物合成和苯丙酸类化合物的生物合成等6条代谢途径。关键酶基因的RT-qPCR结果与黄酮类化合物的相对含量变化趋势基本一致。结论 不同产地雪菊的黄酮代谢物含量和种类差异显著, 可通过检测特定的黄酮类化合物进行雪菊产地的鉴定。 |

Objective To compare and analyze the differences in flavonoid compound composition and key enzyme genes expression between 2 varieties of Coreopsis tinctoria. Methods The flavonoid metabolites in Coreopsis tinctoria were analyzed and identified based on the method of ultra performance liquid chromatography- tandem quadrupole time-of-flight mass spectrometry, while the expression differences of key enzyme genes for flavonoid synthesis were analyzed by quantitative real-time PCR (RT-qPCR). Results A total of 204 flavonoid components were analyzed and identified from Coreopsis tinctoria, and 15 kinds of differentially expressed flavonoid metabolites were screened out, among which 6 kinds of flavonoids were higher in plain Coreopsis tinctoria and 9 kinds of flavonoids were higher in plateau Coreopsis tinctoria. Pathway enrichment analysis of differential metabolites revealed that poor isoflavone metabolites were significantly enriched in 6 metabolic pathways, including flavonoid biosynthesis, biosynthesis of secondary metabolites, isoflavone biosynthesis and biosynthesis of phenylpropanoids. RT-qPCR results of key enzyme genes were generally consistent with the relative contents of flavonoids. Conclusion The types and content of flavonoid metabolites of Coreopsis tinctoria from different origins vary significantly, and the identification of Coreopsis tinctoria origins can be carried out by detecting specific flavonoid compounds. |
