目的 研究星鳗抗氧化肽(Astroconger myriaster antioxidant peptide, CMAPⅠ)在加工贮藏及胃肠消化过程中的稳定性。方法 以1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl, DPPH)自由基清除活性和羟自由基(hydroxyl free radicals, ·OH)清除活性为指标, 通过测定温度、pH、冻融次数、紫外照射时间、不同食品辅料(NaCl、葡萄糖、苹果酸)和不同金属离子对CMAPⅠ稳定性的影响, 同时以体外模拟胃肠消化系统来测定其消化吸收稳定性。结果 CMAPⅠ耐热性较好, 温度在60~80℃时DPPH自由基和·OH清除率分别为71.2%和47.8%。CMAPⅠ抗碱性较差, 当pH为10时, 清除率降低了23.5%和16.6%, 在反复冷冻解冻和紫外照射下对CMAPⅠ的活性基本没有产生影响。添加NaCl、葡萄糖溶液、苹果酸后, 有利于DPPH自由基和·OH清除率的提高。K+和Ca2+对抗氧化活性基本不产生影响, 但随着Mg2+、Cu2+、Fe3+金属离子浓度的增大, CMAPⅠ的活性会明显降低。在体外模拟胃消化时间为60 min时, DPPH自由基和·OH清除率最高, 分别为81.24%和62.28%, 同时, CMAPⅠ在胃中始终能表现较好的稳定性, 而进一步转向肠消化后, 在肠消化300 min时, DPPH自由基和·OH清除率分别为69.08%和46.18%, 肠道消化前后相比降低了12.0%和17.2%。结论 研究表明CMAPⅠ具有一定的抗氧化稳定性, 对CMAPⅠ的工业生产和应用具有重大意义。 |

Objective To study the stability of Astroconger myriaster antioxidant peptide (CMAPⅠ) during processing, storage and gastrointestinal digestion. Methods The effects of temperature, pH, number of freeze-thaws, ultraviolet irradiation time, different food excipients (NaCl, glucose, malic acid) and different metal ions on the stability of CMAPⅠ were measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and hydroxyl free radicals (·OH) scavenging activity as indicators, and the digestive and absorption stability was also determined by simulating the gastrointestinal digestive system in vitro. Results CMAPⅠ had better heat resistance, the DPPH radicals and ·OH scavenging rates were 71.2% and 47.8%, respectively at 60-80℃. CMAPⅠ had poor alkaline resistance, and the scavenging rate decreased by 23.5% and 16.6% when the pH was 10. There was basically no effect on the activity of CMAPⅠ under repeated freezing and thawing and ultraviolet irradiation. The addition of NaCl, glucose solution and malic acid facilitated the increase of DPPH radicals and ·OH scavenging rates. K+ and Ca2+ antioxidant activities were basically unaffected, but Mg2+, Cu2+ and Fe3+ decreased significantly with increasing metal ion concentrations in CMAPⅠ. During in vitro simulated gastric digestion, the highest DPPH radical and ·OH scavenging rates of 81.24% and 62.28%, respectively, were observed at a digestion time of 60 min, while CMAP I always showed good stability in the stomach, while after further transfer to intestinal digestion, the DPPH radical and ·OH scavenging rates were 69.08% and 46.18%, respectively, at 300 min of intestinal digestion, the DPPH radical and ·OH scavenging rates were 69.08% and 46.18%, respectively, and decreased by 12.0% and 17.2% before and after intestinal digestion, respectively. Conclusion The study showes that CMAPⅠ has certain antioxidant stability, which is of great significance to the industrial production and application of CMAPⅠ. |
