目的 建立一种快速的超高效液相色谱(ultra performance liquid chromatography, UPLC)-二极管阵列变波长检测方法, 在10 min内同时测定糕点中安赛蜜、糖精钠、苯甲酸、山梨酸、脱氢乙酸和纳他霉素6种食品添加剂。方法 样品经20%甲醇水溶液(含2 mmol/L甲酸)提取, 乙酸锌、亚铁氰化钾沉淀蛋白, 以甲醇和20 mmol/L乙酸铵溶液(含2 mmol/L甲酸)作为流动相梯度洗脱, 使用Agilent Poroshell 120 EC-C18 (100 mm× 4.6 mm, 2.7 μm)色谱柱分离, 二极管阵列-变波长检测。结果 6种食品添加剂在0.1~100.0 μg/mL范围内线性关系良好, 相关系数均大于0.999, 回收率为83.0%~110.2%, 相对标准偏差为0.56%~5.96%。安赛蜜、糖精钠、苯甲酸、山梨酸、脱氢乙酸和纳他霉素的检出限为0.11~0.29 mg/kg, 定量限为0.38~0.97 mg/kg。结论 该方法前处理简单, 分析时间短, 重现性好, 适合用于糕点中6种食品添加剂的大批量检测。 |

Objective To establish a rapid detection method of ultra performance liquid chromatography (UPLC)-diode array with variable wavelength, and simultaneously determine 6 kinds of food additives, including acesulfame potassium, sodium saccharin, benzoic acid, sorbic acid, dehydroacetic acid and natamycin in pastries within 10 minutes. Methods Samples were extracted by 20% methanol aqueous solution (containing 2 mmol/L formic acid), protein was precipitated by zinc acetate and potassium ferrocyanide, methanol and 20 mmol/L ammonium acetate solution (containing 2 mmol/L formic acid) were used as mobile phase gradient elution, and Agilent Poroshell 120 EC-C18 (100 mm×4.6 mm, 2.7 μm) column was used for separation, and diode array-wavelength-changing detection was used. Results The linear relationships of the 6 kinds of food additives were good within the range of 0.1?100.0 μg/mL, with correlation coefficients greater than 0.999, recoveries of 83.0%?110.2%, and relative standard deviations of 0.56%?5.96%. The limits of detection for acesulfame potassium, sodium saccharin, benzoic acid, sorbic acid, dehydroacetic acid and natamycin were 0.11?0.29 mg/kg, and the limits of quantitation were 0.38?0.97 mg/kg. Conclusion This method has the advantages of simple pretreatment, short analysis time and good reproducibility, and is suitable for mass detection of 6 kinds of food additives in pastry. |
