      目的 了解铜绿假单胞菌的分子多样性和耐药性, 采用16S rRNA基因序列分析法对2018—2020年间分离自广东地区桶装水中的61株铜绿假单胞菌进行同源性分析, 并探讨其对11种不同抗生素的耐药性。方法 采用27F/1492R通用引物对目标菌的16S rRNA基因序列扩增, 测序后, 采用Clustal X软件进行序列比对, 利用MEGA 7.0软件构建系统发生树; 药敏实验应用微生物全自动鉴定及药敏系统检测。结果 系统发育分析结果表明: 61株共可分为3个分支, 分别包含16株一支、1株一支、44株和铜绿假单胞菌标准菌株ATCC 27853一支; 药敏结果显示61株铜绿假单胞菌处于较低耐药水平, 对所有测试抗生素的耐药率均低于2%, 但仍发现一株6重耐药菌株。结论 本研究初步建立了广东地区水源性铜绿假单胞菌的菌株资源库及药敏数据库, 为之后的污染溯源工作提供了数据支持, 从而保障广大消费群众的饮水安全。

      Objective To investigate the molecular diversity and antibiotic resistance of Pseudomonas aeruginosa, analyze the homology of 61 strains of Pseudomonas aeruginosa isolated from bottled water in Guangdong Province from 2018 to 2020 by 16S rRNA gene sequence analysis, and explore their drug resistance to 11 different antibiotics. Methods The 16S rRNA gene sequence of the target bacteria were amplified by 27F/1492R, PCR products were sent for sequencing, and sequence alignment was performed by ClusterX software, and the phylogenetic tree was constructed by MEGA 7.0 software. And the drug sensitivity test was performed by VITEK 2 compact system. Results Phylogenetic analysis showed 61 strains of bacteria could be divided into 3 branches: Sixteen strains formed a branch, one strain formed a branch, the other 44 strains formed a separate branch with Pseudomonas aeruginosa ATCC 27853. The results of drug sensitivity showed that 61 strains of Pseudomonas aeruginosa were in a low level of drug resistance, the resistance rate to all tested antibiotics was less than 2%, despite this, a 6-fold resistant strain was found. Conclusion This study preliminarily establishes the resource database and drug sensitivity database of Pseudomonas aeruginosa isolated from Guangdong, it provides data supports for the pollution traceability of Pseudomonas aeruginosa in barreled water, thus ensuring the safety of drinking water for the majority of consumers.
