目的 研究菊花三七胶囊、菊花提取物及三七提取物对D-半乳糖衰老模型小鼠的抗氧化作用。方法 通过腹腔注射5%的D-半乳糖连续6周, 建立小鼠亚急性衰老模型。不同剂量给药结束后, 检测小鼠血清、脑组织、肝脏匀浆液及皮肤组织中相关抗氧化指标; 计算小鼠的脾脏等脏器指数。结果 与模型组比较, 给药各剂量组能升高模型动物血清和肝脏匀浆液中超氧化物歧化酶、总抗氧化能力、过氧化氢酶、谷胱甘肽过氧化物酶活力及还原性谷胱甘肽含量, 脑组织中超氧化物歧化酶、谷胱甘肽过氧化物酶活力及还原性谷胱甘肽含量, 降低丙二醛含量(P<0.05或P<0.01), 可升高模型动物皮肤组织中超氧化物歧化酶、三磷酸腺苷、过氧化氢酶活性及羟脯氨酸、Ⅰ型胶原蛋白、Ⅲ型胶原蛋白的含量, 降低脂褐素含量(P<0.05或P<0.01), 升高模型小鼠脾脏、肾脏指数, 降低肝脏指数(P<0.05或P<0.01), 且菊花三七胶囊组的多项指标均优于其他剂量组, 菊花提取物高剂量组的多项抗氧化指标优于菊花提取物低剂量组和三七提取物高、低剂量组; HE染色结果表明, 各剂量组小鼠的皮肤和肝脏组织因D-半乳糖损伤所致的组织病理改变程度较模型组均有所减轻, 其中菊花三七胶囊的改善程度最为明显, 基本接近空白对照组; 急性毒性试验表明, 在观察期内各组小鼠无明显中毒症状, 无死亡, 体重增长正常, 解剖未见明显异常。结论 菊花三七胶囊、菊花提取物及三七提取物对小鼠皮肤衰老模型具有一定保护作用, 具有一定的抗氧化功能, 且菊花提取物和三七提取物具有协同增效的作用。 |

Objective To study the antioxidant effects of Chrysanthemum-Panax notoginseng capsule, Chrysanthemum extract and Panax notoginseng extract on D-galactose aging model mice. Methods Subacute aging model of mice was established by intraperitoneal injection of 5% D-galactose for 6 weeks. After administration of different doses, related antioxidant indexes in serum, brain tissue, liver homogenate and skin tissue of mice were detected; the index of spleen and other organs was calculated. Results Compared with model group, the activities of superoxide dismutase, total antioxidant capacity, catalase, glutathione peroxidase and content of glutathione in serum and liver homogenate, superoxide dismutase, glutathione peroxidase and content of glutathione in brain tissue of model animals were increased in each dose group, while the content of malondialdehyde was decreased (P<0.05 or P<0.01), increased the activities of superoxide dismutase, adenosine triphosphate and catalase and the contents of hydroxyproline, type Ⅰ collagen and type Ⅲ collagen in skin tissues of model animals, and decreased the content of lipofuscin (P<0.05 or P<0.01), the spleen and kidney indexes of model mice were increased, and the liver indexes were decreased (P<0.05 or P<0.01), the antioxidant indexes of Chrysanthemum-Panax notoginseng capsule group were better than other dose groups; the antioxidant indexes of Chrysanthemum extract high-dose group were better than those of Chrysanthemum extract low-dose group and Panax notoginseng high-dose and low-dose group; HE staining results showed that the degree of histopathological changes caused by D-galactose injury in the skin and liver tissues of mice in all dose groups was less than that in the model group, and the improvement degree of Chrysanthemum-Panax notoginseng capsule was the most obvious, which was almost close to the blank control group; acute toxicity test showed that during the observation period, mice in each group had no obvious poisoning symptoms, no death, normal weight growth, no obvious abnormalities in anatomy. Conclusion Chrysanthemum-Panax notoginseng capsule, Chrysanthemum extract and Panax notoginseng extract have certain protective effect on mouse skin aging model, and have certain antioxidant function, and Chrysanthemum extract and Panax notoginseng extract have synergistic effect. |
