      目的 研究苹果渣中粗多糖(apple pomace polysaccharides, AP)对链脲佐菌素(streptozotocin, STZ)诱导的糖尿病大鼠的降血糖和降血脂活性。方法 考察AP对α-葡萄糖苷酶和α-淀粉酶活性的抑制, 研究其体外降糖作用。用不同剂量AP治疗链脲佐菌素诱导的糖尿病大鼠, 测定其大鼠体重、食物和水摄入量及肝脏和骨骼肌糖原含量、血脂水平。结果 当口服800 mg/kg AP 6周后, 糖尿病大鼠的食物和水摄入量以及空腹血糖和胰岛素水平显著降低, 肝脏和骨骼肌糖原含量、丙酮酸激酶和己糖激酶浓度显著升高(P<0.01或P<0.05)。总胆固醇(total cholesterol, TC)、甘油三酯(triglyceride, TG)、低密度脂蛋白胆固醇(low-density lipoprotein-cholesterol, LDL-C)和极低密度脂蛋白胆固醇(very low-density lipoprotein-cholesterol, VLDL-C)水平显著降低, 而高密度脂蛋白胆固醇(high-density lipoprotein-cholesterol, HDL-C)水平显著升高(P<0.01或P<0.05), 其可改善糖尿病大鼠的氧化应激水平。结论 苹果渣多糖对糖尿病SD大鼠具有明显的降血糖、降血脂效果, 其可作为辅助性治疗糖尿病的功能性食品成分。

      Objective To study the hypoglycemic and hypolipidemic activities of apple pomace polysaccharides (AP) on diabetic SD rats induced by streptozotocin (STZ). Methods The effects of AP on α-glucosidase and α-amylase inhibitory were conducted to study its hypoglycemic effect in vitro. Different doses of AP were used to treat STZ induced diabetic mice. The body weight, food and water intake, liver and skeletal muscle glycogen content and blood lipid level were determined. Results After orally administrated with 800 mg/kg AP for 6 weeks, the food and water intake and the levels of fasting blood glucose and insulin in blood for the diabetic rats were drastically decreased, while the content of liver and skeletal muscle glycogen and the concentrations of hepatic pyruvate kinase and hexokinase were obviously increased (P<0.01 or P<0.05). The levels of total cholesterol (TC), triglyceride (TG), low-density lipoprotein-cholesterol (LDL-C) and very low-density lipoprotein-cholesterol (VLDL-C) were significantly descended, while high-density lipoprotein-cholesterol (HDL-C) was significantly ascended (P<0.01 or P<0.05). In addition, it could improve the level of oxidative stress in diabetes rats. Conclusion AP has obvious hypoglycemic and hypolipidemic effects on diabetes SD rats. It can be used as a functional food component for the adjuvant treatment of diabetes.
