目的 优化巴西莓原花青素的提取工艺, 并研究其抗疲劳功效。方法 采用超声和溶剂萃取法相结合提取原花青素, 通过单因素及响应面分析优化工艺参数。建立小鼠运动疲劳模型, 选用ICR雄性小鼠80只, 分为对照组、低剂量组、中剂量组、高剂量组, 对照组喂基础饲料, 实验组灌胃不同剂量的巴西莓提取物, 分别为1.7、16.8、50.0 mg/kg体重, 实验周期为28 d, 记录小鼠游泳力竭时间, 测定运动后血乳酸、尿素氮、肝糖原及肌糖原含量。结果 经优化验证后, 最佳工艺条件为乙醇浓度70%、料液比1:20 (g:mL)、超声功率180 W、超声温度75 ℃、超声时间50 min, 原花青素提取率达2.0278%。各剂量组小鼠负重游泳时间均比对照组延长, 高剂量组具有极显著差异(P<0.01); 各剂量组小鼠运动后血乳酸含量均低于对照组, 肝糖原、肌糖原含量均高于对照组。结论 优化后的提取条件能够有效提取巴西莓中的原花青素, 且巴西莓原花青素具有抗疲劳作用。 |

Objective To optimize the extraction process of acai berry proanthocyanidins and study its anti-fatigue effect. Methods The proanthocyanidins were extracted by a combination of ultrasound and solvent extraction, and the process parameters were optimized by single factor and response surface analysis. The mouse exercise fatigue model was established, 80 ICR male mice were selected and divided into control group, low-dose group, middle-dose group, and high-dose group. The control group was fed with basic feed, and the experimental groups were given different doses of acai berry extracts of 1.7, 16.8, and 50.0 mg/kg body weight, respectively. The experimental period was 28 d, the swimming exhaustion time of mice was recorded, and the content of blood lactate, blood urea nitrogen, liver glycogen and muscle glycogen after exercise were determined. Results After optimization and verification, the optimal process conditions were ethanol concentration 70%, material-liquid ratio 1:20 (g:mL), ultrasonic power 180 W, ultrasonic temperature 75 ℃, ultrasonic time 50 min, and the extraction rate of proanthocyanidins reached 2.0278%. The weight-bearing swimming time of mice in each dose group was longer than that in the control group, and there was a very significant difference in the high-dose group (P<0.01). The blood lactate content of mice in each dose group after exercise was lower than that of the control group, and the liver glycogen and muscle glycogen content were higher than those of the control group. Conclusion The optimized extraction conditions can effectively extract proanthocyanidins from acai berry, and acai berry proanthocyanidins have anti-fatigue effect. |
