目的 研究川牛膝(Cyathulae radix)水提物对环磷酰胺(cyclophosphamide, CTX)诱导的免疫抑制小鼠免疫功能的影响。方法 雌性ICR小鼠60只, 根据体重随机分为空白对照组、免疫抑制模型组和川牛膝低、中、高干预剂量组, 每组12只。实验第1~3 d, 除对照组外, 其余各组小鼠每天腹腔注射80 mg/(kg·bw)环磷酰胺, 建立免疫抑制小鼠模型, 对照组每天腹腔注射等量生理盐水。第4 d起, 每天给予对照组和模型组1%羧甲基纤维素钠(carboxy methylcellulose sodium, CMC-Na)溶液灌胃, 川牛膝低、中、高剂量组分别给予0.06、0.12、0.24 g/(kg·bw)川牛膝提取物的CMC-Na混悬液灌胃, 连续灌胃30 d后测定小鼠体重、脾脏和胸腺质量及形态、外周血白细胞计数、免疫球蛋白含量、细胞因子水平、外周血及脾脏T淋巴细胞亚群、骨髓有核细胞计数和DNA含量。结果 川牛膝水提物3个干预剂量组小鼠与模型组小鼠相比: 脾脏和胸腺的形态学结构逐渐恢复; 外周血白细胞计数不同程度的增加; 免疫球蛋白IgG含量增加, IgM含量在低、高2个剂量组显著增加; 血清干扰素-γ (interferon-γ, IFN-γ)水平在高剂量组显著升高, 肿瘤坏死因子-α (tumor necrosis factor-α, TNF-α)浓度在低剂量组恢复, 白细胞介素-2 (interleukin-2, IL-2)浓度在低剂量组略有升高, 在中、高剂量组显著下降, IL-9浓度在高剂量组有所降低, 且较其他组向对照组回归程度最大, IL-10浓度显著降低; 外周血CD3+、CD4+、CD8+ T淋巴细胞百分比在低剂量组显著恢复, 脾脏CD3+、CD4+ T淋巴细胞百分比在3个组均显著恢复, 且恢复程度随着川牛膝剂量的提高而提高, CD8+ T淋巴细胞百分比在低、高剂量组显著恢复; 骨髓有核细胞浓度在低、高剂量组有显著提高, DNA含量在高剂量组显著增加。结论 一定剂量的川牛膝水提物对环磷酰胺诱导的免疫抑制小鼠的免疫功能具有改善作用。 |

Objective To evaluate the effects of water extract of Cyathulae radix on the immune function in immunosuppressed mice induced by cyclophosphamide. Methods Sixty female ICR mice were randomly divided into control, immunosuppression model and low-, medium- and high-dose Cyathulae radix intervention groups, with 12 mice in each group. In the first 3 days of the experiment, the mice in all groups except the control group were intra peritoneally injected with cyclophosphamide at a daily dose of 80 mg/(kg·bw) to establish an immunosuppressed mouse model, and the mice in the control group were intra peritoneally injected with the same amount of normal saline every day. From the 4th day, the mice in the control and cyclophosphamide model group were orally administered with 1% carboxy methylcellulose sodium (CMC-Na) solution, and those in the low-, medium- and high-dose groups of Cyathulae radix were administered with Cyathulae radix at doses of 0.06, 0.12, 0.24 g/(kg·bw), after continuous gavage for 30 days, the body weight, the weight and morphology of spleen and thymus, white blood cell count, immunoglobulin content and cytokine levels in peripheral blood, peripheral blood and spleen T lymphocyte subsets, bone marrow nucleated cells count and DNA content were measured. Results Compared with the model group, 3 Cyathulae radix intervention groups: The morphological structure of the spleen and thymus tissues gradually recovered; white blood cell count in peripheral blood increased to varying degrees; the content of immunoglobulin IgG increased, and the content of IgM increased significantly in low- and high-dose groups; the serum interferon-γ (IFN-γ) concentration increased significantly in the high-dose group, and the tumor necrosis factor-α (TNF-α) concentration recovered in the low-dose group, and the concentration of interleukin-2 (IL-2) increased slightly in the low-dose group, and decreased significantly in the medium- and high-dose groups, while the IL-9 concentration decreased in the high-dose group, and the degree of regression to the control group was the largest compared with other groups, IL-10 concentration decreased significantly; the percentages of CD3+, CD4+ and CD8+ T lymphocytes in peripheral blood significantly recovered in the low-dose group, while the percentages of CD3+ and CD4+ T lymphocytes in spleen significantly recovered in the 3 groups, and the degree of recovery gradually increased with the dose of Cyathulae radix, and the percentage of CD8+ T lymphocytes in spleen significantly recovered in the low- and high-dose groups; the concentrations of bone marrow nucleated cells were significantly increased in the low- and high-dose groups, and the DNA content was significantly increased in the high-dose group. Conclusion A certain dose of water extract of Cyathulae radix can improve the immune function of immunosuppressed mice induced by cyclophosphamide. |
