目的 建立高效液相色谱法(high performance liquid chromatography, HPLC)同时检测食品中9种合成着色剂(柠檬黄、新红、苋菜红、靛蓝、胭脂红、日落黄、诱惑红、亮蓝及酸性红)的分析方法。方法 样品经超声提取、聚酰胺净化、浓缩、定容、过膜, 经Shim-pack GIST C18色谱柱(250 mm×4.6 mm, 5 μm)分离, 以甲醇和0.02 mol/L乙酸铵为流动相, 梯度洗脱分离进二极管阵列检测器检测。结果 在0.4~20.0 μg/mL浓度范围内, 线性关系良好, 相关系数均在0.999以上, 方法检出限为0.5 mg/kg; 分别对多种样品进行低 (0.5 mg/kg)、中(1.0 mg/kg)、高(5.0 mg/kg) 3个浓度加标回收率实验, 回收率范围为76.2%~107.1%, 相对标准偏差(relative standard deviations, RSDs)为0.4%~8.5% (n=6)。结论 该方法分析速度快、准确度高、重复性好, 适用于各种基质食品中9种合成着色剂的同时检测。 |

Objective To establish a method for the simultaneous determination of 9 kinds of synthetic colorants (lemon yellow, new red, amaranth, indigo, carmine, sunset yellow, temptation red, brilliant blue and acid red) in foods by high performance liquid chromatography (HPLC). Methods After ultrasonic extraction, polyamide purification, concentration, constant volume, membrane separation, the samples were separated on a Shim-pack GIST C18 column (250 mm×4.6 mm, 5 μm) using methanol and 0.02 mol/L ammonium acetate as the mobile phase, and detected by a diodearray detector after gradient elution separation. Results Within the concentration range of 0.4 to 20.0 μg/mL, the linear relationship was good and the correlation coefficients were all above 0.999. The limits of detection of this method were 0.5 mg/kg, the recoveries of various samples were tested at low (0.5 mg/kg), medium (1.0 mg/kg) and high (5.0 mg/kg), the recoveries ranged from 76.2% to 107.1%, and the relative standard deviations (RSDs) were 0.4% to 8.5% (n=6). Conclusion This method has the advantages of fast analysis speed, high accuracy and good repeatability, whichis suitable for the simultaneous determination of 9 kinds of synthetic colorants in various matrix foods. |
