目的 优化超声辅助法提取牡丹花中总黄酮和总多酚的工艺。方法 在单因素的基础上, 采用响应面实验设计, 以总黄酮和总多酚含量总评值“归一值(overall desirability, OD)”为最终响应值, 分别考察乙醇浓度、料液比、提取时间、提取温度4个因素对总黄酮和总多酚得率的影响。结果 4个因素对牡丹花中2种活性成分的提取均有显著影响, 响应面优化工艺为: 料液比1:36 (m:V)、乙醇浓度60%、提取时间43 min、提取温度50 ℃。在此条件下, 总黄酮提取率为1.39%, 总多酚提取率为3.20%, 预测OD为0.95。结论 所建模型能够较好地预测牡丹花总黄酮和总多酚的提取率, 本研究为牡丹花的综合开发提供了理论依据。 |

Objective To optimize the extraction process of total flavonoids and total polyphenols from peony flowers by ultrasonic assisted method. Methods On the basis of single factor, the response surface design was adopted, and the total evaluation value “overall desirability (OD)” of total flavonoids and total polyphenols was taken as the final response value, the effects of ethanol concentration, solid-liquid ratio, extraction time and extraction temperature 4 factors on the yields of total flavonoids and total polyphenols were investigated. Results The 4 factors had significant effects on the extraction of 2 kinds of active components from peony flowers, the response surface optimization process was as follows: Solid-liquid ratio 1:36 (m:V), ethanol concentration 60%, extraction time 43 min, extraction temperature 50 ℃. Under these conditions, the extraction rate of total flavonoids was 1.39%, the extraction rate of total polyphenols was 3.20%, and the predicted OD was 0.95. Conclusion The established model can better predict the extraction rate of total flavonoids and total polyphenols frompeony flowers. This study provides a theoretical basis for the comprehensive development of peony. |
