目的 建立高效液相色谱-电雾式检测器法(high performance liquid chromatography-charged aerosol detection, HPLC-CAD)同时测定烘焙食品中果糖、葡萄糖、乳糖、半乳糖、蔗糖和麦芽糖含量的分析方法。 方法 以NH2P-50柱(250 mm×4.6 mm, 5 μm)为分离柱, 乙腈-水(78:22, V:V)为流动相, 等度洗脱, 流速为 0.8 mL/min, 以CAD为检测器, 雾化温度35 ℃。结果 方法在40 min内完成了6种糖的定量分析。果糖、葡萄糖、乳糖和半乳糖在2~200 mg/L、蔗糖和麦芽糖在4~400 mg/L范围内有良好的线性关系(r2>0.999), 相对标准偏差(relative standard deviations, RSDs)不超过3.3%, 回收率不小于71.0%。结论 该方法快速、灵敏、准确度高, 可为烘焙食品中糖类物质的含量测定提供技术支持, 也可以为食品安全监督抽检和居民膳食控制提供科学依据。 |

Objective To establish an analytical method for simultaneous determination of fructose, glucose, lactose, galactose, sucrose and maltose in baked foods by high performance liquid chromatography-charged aerosol detection (HPLC-CAD). Methods NH2P-50 column (250 mm×4.6 mm, 5 μm) was used as the separation column, acetonitrile-water (78:22, V:V) was used as the mobile phase, and isocratic elution was performed at the flow rate of 0.8 mL/min using CAD as the detector and at the atomization temperature of 35 ℃. Results The quantitative analysis of 6 kinds of sugars was completed within 40 min. The linear relationships of fructose, glucose, lactose and galactose were good in the range of 2?200 mg/L, and sucrose and maltose were good in the range of 4? 400 mg/L (r2>0.999), the relative standard deviations (RSDs) were not more than 3.3%, and the recovery rates were not less than 71.0%. Conclusion The method is rapid, sensitive and accurate, which can provide technical support for the determination of sugar content in baked food, as well as scientific basis for food safety supervision and sampling inspection and residents’ dietary control. |
