      目的 优化微波辅助酶法提取葛根黄酮的工艺, 并对葛根黄酮的抗氧化活性进行评价。方法 以葛根为原料, 葛根黄酮得率为考核指标, 在微波辅助、酶催化作用下, 在单因素试验基础上, 利用正交试验法对葛根黄酮提取工艺进行优化, 并对葛根黄酮的抗氧化活性进行研究。结果 最佳提取工艺参数为: 复合酶添加量3.0%、酶解pH=6、微波功率200 W、提取时间40 min, 在此条件下, 葛根黄酮的得率为6.79%。葛根黄酮的抗氧化活性随着质量浓度的增加而增强, 当葛根黄酮的质量浓度为200 μg/mL时, 葛根黄酮对1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl, DPPH)自由基的清除率为80.06%、对羟自由基的清除率为73.28%。结论 正交试验法优化了葛根黄酮的提取工艺, 得到的葛根黄酮具有较强的抗氧化活性。

      Objective To optimize the extraction process of Radix puerariae flavonoids by microwave-assisted enzymatic method, and evaluate the antioxidant activity of flavonoids. Methods Taking Radix puerariae as raw material, the yield of Radix puerariae flavonoids as evaluation index, microwave-assisted, enzyme catalyzed, on the basis of single factor experiment, orthogonal test was used to optimize the extraction process of Radix puerariae flavonoids, and the antioxidant activity of Radix puerariae flavonoids was studied. Results The optimum extraction conditions were as follows: 3.0% compound enzyme, pH=6, microwave power 200 W, extraction time 40 min. Under these conditions, the yield of Radix puerariae flavonoids was 6.79%. The antioxidant activity of Radix puerariae flavonoids increased with the increase of mass concentration, when the mass concentration of Radix puerariae flavonoids was 200 μg/mL, the scavenging rates of Radix puerariae flavonoids on 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals and hydroxyl radicals were 80.06% and 73.28%. Conclusion The extraction process of Radix puerariae flavonoids is optimized by orthogonal test, and the obtained flavonoids have strong antioxidant activity.
