      目的 建立一种快速检测水产品中呋喃唑酮代谢物的荧光免疫层析试纸。方法 以紫外灯下呈鲜亮红色的铕荧光微球为抗体标记物, 通过调试荧光微球-抗体复合物的工作液配方, 优化检测线和控制线, 改善试纸反应性能和荧光显色梯度, 提高试纸灵敏度。结果 结果显示, 通过肉眼判断本试纸对呋喃唑酮代谢物的最低检出限为0.25 ?g/kg, 特异性良好, 在冷藏条件下其稳定性可达1年。在40批次不同品种水产品的检测中, 试纸检测结果与液相色谱-串联质谱法的检测结果符合程度为100%。结论 呋喃唑酮代谢物荧光免疫层析试纸具有快速简单、准确灵敏、成本低的优势, 其结果易于判定, 无需专门的检测仪器, 适用于大规模筛查、多环节质控和风险评估, 具有广阔的应用前景。
英文摘要:
      Objective To develop a fluorescence immunochromatographic strip for the rapid detection furazolidone metabolites in aquatic products. Methods Europium fluorescent microspheres with bright red under an ultraviolet lamp were used as antibody markers, and the detection line and the control line were optimized by debugging the working solution formula of the fluorescent microspheres-antibody complex, so that the reaction performance and the fluorescence color gradient of the test paper were improved, and the sensitivity of the test paper was improved. Results The results showed that the minimum detection limit of furazolidone metabolite by the test paper was 0.25 ?g/kg by naked eye, the specificity was good, and the stability of the test paper could be up to 1 year under refrigeration. In the detection of 40 batches of aquatic products of different varieties, the coincidence degree of the test results with the liquid chromatography-tandem mass spectrometry detection results was 100%. Conclusion Fluorescence immunochromatographic strip for furazolidone metabolites has the advantages of rapidity, simplicity, accuracy, sensitivity and low cost, and its results are easy to judge, without special detection instruments, it is suitable for large-scale screening, multi-link quality control and risk assessment, and has broad application prospects.
