      目的 使用高效液相色谱(high performance liquid chromatography, HPLC)指纹图谱结合指标成分评价湖北虎杖资源。方法 采用高效液相色谱法测定59批不同产地虎杖中虎杖苷、白藜芦醇、大黄素、大黄素-8-O-β-D葡萄糖苷和大黄素甲醚5种有效成分含量, 并用中药指纹图谱相似度评价系统进行相似度分析, 结合聚类分析、主成分分析和偏最小二乘判别分析对其进行质量评价。结果 建立了湖北虎杖的HPLC指纹图谱, 标定了9个共有峰, 经对照品比对指认了5个色谱峰; 相似度和聚类分析显示, 不同地区虎杖质量存在差异, 进一步用主成分分析对各地区虎杖质量进行综合评价, 发现黄冈英山、黄冈蕲春和荆州松滋等地虎杖质量较优; 利用偏最小二乘判别分析确定了白藜芦醇和大黄素-8-O-β-D葡萄糖苷为差异性成分。结论 本研究建立的湖北虎杖HPLC指纹图谱方法快速简便, 可为湖北虎杖规范化和规模化基地建设的优质种质资源选择提供参考。

      Objective To evaluate the resources of Polygonum cuspidatum in Hubei province by high performance liquid chromatography (HPLC) fingerprint combined with index components. Methods The content of 5 kinds of active components including polydatin, resveratrol, emodin, emodin-8-O-β-D glucoside and emodin methyl ether in 59 batches of Polygonum cuspidatum from different origins were determined by high performance liquid chromatography, and the similarity analysis was performed using the similarity evaluation system of Chinese medicine fingerprint chromatograms. The quality was evaluated by combining cluster analysis, principal component analysis and partial least squares discriminant analysis. Results The HPLC fingerprint of Polygonum cuspidatum in Hubei province was established, 9 common peaks were calibrated, and 5 chromatographic peaks were identified by comparison of reference substances. Similarity and cluster analysis showed that there were differences in the quality of Polygonum cuspidatum in different regions. Furthermore, the quality of Polygonum cuspidatum in different regions was comprehensively evaluated by principal component analysis, and it was found that the quality of Yingshan in Huanggang, Qichun in Huanggang and Songzi in Jingzhou were better. Partial least squares discriminant analysis was used to identify resveratrol and emodin-8-O-β-D glucoside as different components. Conclusion The established HPLC fingerprint method of Polygonum cuspidatum in Hubei province is rapid and simple, which can be used as a reference for selecting high-quality germplasm resources for standardization and scale base construction of Polygonum cuspidatum in Hubei province.
