目的 建立一测多评法(quantitative analysis of multi-components by single marker, QAMS)同时测定桑叶中5种黄酮类成分(芦丁、异槲皮苷、紫云英苷、槲皮素、山奈酚)的分析方法。方法 采用高效液相色谱法, 建立其他成分与参照物异槲皮苷的相对校正因子, 通过主成分分析及聚类热图进行聚类分析。结果 相对校正因子具有良好的耐用性, 相对标准偏差(relative standard deviation, RSD)值均小于3%; 一测多评法与外标法所测结果无明显差异(RSD值均小于3.5%); 不同产地桑叶中黄酮类成分含量差异较大, 其中芦丁含量测定结果为0.0104%~0.0529%, 异槲皮苷为0.0747%~0.1594%, 紫云英苷为0.0196%~0.0537%, 槲皮素为0.0004%~0.0009%, 山奈酚为0.0002%~0.0005%, 且无明显产地聚类特性。结论 一测多评法适用于桑叶中5种黄酮类成分的同时测定, 桑叶需规范化种植以保证质量的可靠性, 确保其功效的稳定及安全。 |

Objective To establish a method for the simultaneous determination of 5 kinds of flavonoids (rutin, isoquercitrin, avidin, quercetin, kaempferol) from Morus alba L. by quantitative analysis of multi-components by single marker (QAMS). Methods High performance liquid chromatography (HPLC) was used to establish the relative correction factors of other components with the reference substance isoquercitrin, and cluster analysis was performed by principal component analysis and clustering heat map. Results The relative correction factor had good durability and relative standard deviation (RSD) values were all less than 3%. There was no significant difference between the QAMS method and the external standard method (RSD values were all less than 3%). The content of flavonoids in Morus alba from different producing areas was quite different, and the determination result of rutin content was between 0.0104%?0.0529%, isoquercitrin was between 0.0747%?0.1594%, astragaloside was between 0.0196%?0.0537%, quercetin was between 0.0004%?0.0009%, and kaempferol was between 0.0002%?0.0005%, and there was no obvious clustering characteristics of producing areas. Conclusion The quantitative analysis of multi-components by single marker is suitable for the simultaneous determination of 5 kinds of flavonoids in Morus alba L.. It should be noted that the Morus alba L. need to be planted in a standardized manner to ensure the reliability of the quality, and to ensure the stability and safety of its function. |
