      目的 优化黄芪蛋白水解液工艺条件, 并考察其体外抗氧化活性。方法 在单因素试验的基础上, 选择温度、pH、酶用量、酶解时间为影响因素, 以1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl, DPPH)自由基清除率为指标, 采用中心组合Box-Benhnken法建立数学模型, 进行响应面分析。结果 黄芪水解蛋白的最佳酶解条件为酶解温度50 ℃、pH值3、酶用量800 U/g、酶解时间8 h, 该条件下水解度为52.96%, DPPH自由基清除率为80.64%, 清除率为98.93%, ·OH清除率为74.83%, 还原能力为2.3。结论 黄芪蛋白水解液具有较强的抗氧化活性, 可作为一种天然食品抗氧化剂或抗衰老、抗氧化饮品。

      Objective To optimize the technological conditions of Astragalus membranaceus protein hydrolysate and investigate its antioxidant activity in vitro. Methods On the basis of single factor experiment, temperature, pH, enzyme dosage and enzymatic hydrolysis time were selected as the influencing factors, and 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging rate was taken as the index. The mathematical model for response surface analysis was established by central combination Box-Benhnken method. Results The optimal enzymatic hydrolysis conditions of Astragalus membranaceus protein as follows: Hydrolysis temperature was 50 ℃, pH was 3, enzyme dosage was 800 U/g, hydrolysis time was 8 h. In this conditions, the hydrolysis degree of hydrolyzed protein from Astragalus membranaceus was 52.96%, DPPH radical scavenging rate was 80.64%, and scavenging rate was 98.93%, ·OH scavenging rate was 74.83% and the reduction ability was 2.3. Conclusion Astragalus membranaceus protein hydrolyzate has strong antioxidant activity, which can be used as a natural food antioxidant or anti-aging and antioxidant drink.
