目的 探究黑果枸杞总黄酮抗肝癌作用的可能机制。方法 采用噻唑蓝比色法检测不同浓度黑果枸杞总黄酮对LO-2细胞存活率和HepG-2细胞抑制率的影响, 确定最佳干预时间和浓度, 再以确定的3个质量浓度黑果枸杞总黄酮(25、50和100 μg/mL)和20 μg/mL顺铂给药48 h, 使用细胞仪检测细胞凋亡和周期变化, 通过检测细胞凋亡相关酶的活性, 初步考察黑果枸杞总黄酮抗肝癌的作用机制。结果 黑果枸杞总黄酮对HepG-2细胞的增殖有一定的抑制作用, 抑制率可达98.27%。黑果枸杞总黄酮诱导HepG-2肝癌细胞周期阻滞于S期, 使细胞停止分裂和增殖, 同时可通过上调Bcl-2相关X蛋白(Bcl-2 associated X protein, Bax)和半胱氨酸蛋白酶3 (cysteine proteinase-3, Casp-3)及下调B细胞淋巴瘤因子2 (B-cell leukemia-2, Bcl-2)的表达水平来诱导HepG-2肝癌细胞的凋亡。结论 黑果枸杞总黄酮对肝癌细胞具有一定的抑制作用。本研究可为后期进一步研究黑果枸杞抗肝癌作用机制提供依据。 |

Objective To explore the possible mechanism of total flavonoids of Lycium ruthenicum Murray against liver cancer. Methods Thiazolium blue colorimetry was used to detect the effects of different concentrations of total flavones of Lycium ruthenicum Murray on the survival rate of LO-2 cells and the inhibition rate of HepG-2 cells. The optimal intervention time and concentration were determined, and then the 3 determined concentrations of total flavones of Lycium ruthenicum Murray (25, 50 and 100 μg/mL) and 20 μg/mL cisplatin were administered for 48 h. Cell apoptosis and cycle changes were detected by cell analyzer. The mechanism of action of total flavones of Lycium ruthenicum Murray against liver cancer was preliminarily investigated by detecting the activities of apoptosis-related enzymes. Results The total flavones of Lycium ruthenicum Murray could inhibit the proliferation of HepG-2 cells, and the inhibition rate was up to 98.27%. Total flavones of Lycium ruthenicum Murray induced the arrest of HepG-2 cells cycle in the S phase, which stopped cell division and proliferation, and induced the apoptosis of HepG-2 cells by up-regulating Bcl-2 associated X protein (Bax) and cysteine proteinase-3 (Casp-3) and down-regulating B-cell leukemia-2 (Bcl-2) expression levels. Conclusion The total flavones from Lycium ruthenicum Murray have a certain inhibitory effect on hepatocellular carcinoma cells. This research can provide the basis for further study of the mechanism of Lycium ruthenicum Murray against liver cancer. |
