      目的 建立QuEChERS-超高效液相色谱-串联质谱法(ultra performance liquid chromatography-tandem mass spectrometry, UPLC-MS/MS)同时检测红酒中组胺、酪胺、色胺和苯乙胺4种生物胺的分析方法。方法 红酒样品经改进的QuEChERS混合填料净化后, 经ACQUITY UPLC? BEH Amide色谱柱(2.1 mm×100 mm, 1.7 μm)分离, 以0.1%(V:V)甲酸-15 mmol/L乙酸铵水溶液和乙腈为流动相进行梯度洗脱, 质谱(ESI+)采用多反应监测模式(multiple response monitoring, MRM), 外标法定量。结果 组胺、酪胺、色胺和苯乙胺4种目标化合物在色谱柱上有很好的保留, 在1.00~800 μg/L范围内具有良好线性(r2>0.9995), 4种目标化合物的检出限为1.0 μg/L, 定量限为3.0 μg/L。4种化合物的回收率范围为94.7%~106.7%, 相对标准偏差为3.7%~8.1%。结论 该方法简单、快速、结果准确、灵敏度高, 适合测定红酒中组胺、酪胺、色胺和苯乙胺4种生物胺。

      Objective To establish a method for simultaneous determination of 4 kinds of biogenic amines in wine by QuEChERS-ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS). Methods Modified QuEChERS method was adopted for the purification, the sample solution was separated on an amide column ACQUITY UPLC? BEH Amide (2.1 mm×100 mm, 1.7 μm) using 0.1%(V/V) formic acid-15 mmol/L ammonium acetate and acetonitrile as mobile phase for gradient elution, detected by MS/MS (ESI+) with multiple reactions monitoring (MRM), and quantified by external standard method. Results In this method, 4 target compounds of histamine, tyramine, tryptamine and phenylethylamine were well retained on the chromatographic column with good linearity in the range of 1.00-800 μg/L (r2> 0.9995). The limits of detection and limits of quantification were 1.0 μg/L and 3.0 μg/L. The recoveries of the 4 target compounds ranged from 94.7% to 106.7% with relative standard deviations of 3.7%-8.1%. Conclusion This method is easy, fast, accurate and sensitive, which is suitable for the determination of 4 biogenic amines in wine.
